复合血虚模型中环磷酰胺剂量的探索

摘　　要：目的:研究复合血虚模型中环磷酰胺剂量对血虚证模型及免疫系统的影响。方法:110只小鼠随机分为11组,即正常组、模型组、模型给药组,模型组及模型给药组小鼠分别于实验的第1、4天皮下注射乙酰苯肼生理盐水溶液,剂量分别为20、10mg/kg,第4、5、6、7天腹腔注射环磷酰胺生理盐水溶液,模型1组至5组的注射剂量分别为10 mg/kg、20 mg/kg、30 mg/kg、40 mg/kg、50 mg/kg。其中第4天先皮下注射乙酰苯肼生理盐水溶液,2 h后再腹腔注射环磷酰胺生理盐水溶液,整个模型建立过程共7天,造模的同时给药,各给药组均灌胃给予当归补血汤。结果:通过考察外周血常规、脏器指数、促红细胞生成素(EPO)水平等指标发现环磷酰胺剂量为20 mg/kg时为最佳模型,大剂量的环磷酰胺确实会产生免疫抑制作用,而小剂量环磷酰胺对免疫系统的损伤在给予补血药物后可以得到缓解。结论:本研究为血虚模型中环磷酰胺的用量提供了新的参考。Objective:To investigate the effect of cyclophosphamide on different dosage in Xue Xu model and immune system.Methods:110mice were randomly divided into 11groups :blank control group,Model 1-Model 5group,and Danggui Buxue Decoction 1-Danggui Buxue Decoctian 5group.Except blank control group,all other groups were subcutaneously injectexi acetyl phenyl hydrazine saline solution at dose of 20mg/kg and 10mg/kg on the first day and fourth day,then was intraperitoneal injected on 4th,5th,6th and 7th days,on the dose of 10, 20,30,40,50mg/kg separately to rats in model groups 1to 5.On 4th day,it was important to place aeetyl phenyl hydrazine saline solution injection before eyelophosphamlde saline solution.During the 7days of model.establlshment,Danggui Buxue Decoction was intragastric ad- ministered.In the experiment,peripheral blood ceils,spleen index,thymus index,EPO level and bone marrow pathology were detected. Results:It was suggested that cyclophosphamide at the dose of 20mg/kg was"best on Xue Xu model establishment .High-dose eyclophos- phamide"would exert iramunosuppressive effects and damage,and low-dose cyclophosphamide on immune system might be alleviated by giving blood supplement medicine.Conclusion:This study provides a new reference for cyelophosphamide dosage of Xue Xu model.
