摘 要:目的观察糖尿病大鼠脑缺血再灌注给予七氟醚后处理的影响。方法选取本院动物研究中心提供的清洁级雄性SD大鼠60只,将其随机分为3组,七氟醚后处理组(S组)、脑缺血再灌注组(I/R组)及假手术组(C组),各20只。建立大鼠大脑中动脉阻断再灌注模型(MCAO/R),进行相应的处理,在脑缺血再灌注后的第1天、第2天时,对比分析3组神经功能评分及脑组织内的兴奋性氨基酸天冬氨酸(Asp)及谷氨酸(Glu)的含量。结果在各个时间点C组的兴奋性氨基酸Asp和Glu含量无显著变化,但S组明显高于C组,显著低于I/R组,差异具有统计学意义(P <0. 05),在脑缺血再灌注后的第1天、第2天时I/R组Asp和Glu含量比C组明显增多,差异具有统计学意义(P <0. 05);在各个时间点C组的神经功能评分无显著变化,I/R组与S组神经功能评分均高于C组,但I/R组明显高于S组,差异具有统计学意义(P <0. 05)。结论在糖尿病大鼠脑缺血再灌注中应用七氟醚后处理,可明显减少大鼠脑组织中兴奋性氨基酸Glu和Asp的含量,脑缺血再灌注损伤显著减轻,有效保护神经功能,从而改善预后。Objective To explore effects of sevoflurane post-condioning on cerebral ischemia-reperfusion in diabetic rats. Methods To select sixty maleness rats and divide the rats into three groups randomly( n = 20),sevoflurane post-condioning group( group S),cerebral ischemia-reperfusion group( group I/R),contrast operation group( group C).To set up models of ischemia-reperfusion through blocking the middle cerebral artery of rats,and contrast the score of nerves and the content of Asp and Glu of brain tissue in three groups at 1 d and 2 d after cerebral ischemia-reperfusion.Results The content of Asp and Glu in group C had no significant change at each time point,however,group S was higher than group C and lower than group I/R obviously,which differences had statistical significance( P < 0. 05). The content of Asp and Glu in group I/R was higher than group C obviously at 1 d and 2 d after cerebral ischemia reperfusion which differences had statistical significance( P < 0. 05). The score of nerves in group C had no significant change at each time point,however,group I/R and group S were higher than group C and group I/R was higher than group S obviously which differences had statistical significance( P < 0. 05). Conclusion Sevoflurane post-condioning on cerebral ischemia-reperfusion in diabetic rats could reduce the content of Asp and Glu of brain tissue obviously,relieve the cerebral ischemia-reperfusion injury,and preserve the function of nerves effectively thus improving prognosis.
