摘  要:利用小麦秸秆和甘蔗渣制备木质纤维素水解液,确立了可行的脱毒方法,研究酵母菌株Trichosporon dermatis ZZ-46在脱毒水解液与未脱毒水解液中的发酵产油能力,并采用摇瓶发酵培养,对菌株发酵过程、产油能力、发酵动力学以及油脂脂肪酸组成进行了探索。结果表明:采用氢氧化钙与活性炭混合脱毒法最佳,此时糠醛毒性物质含量最低,仅为0.032 2μg/mL,ZZ-46利用该脱毒水解液发酵产油较高,120 h时为ZZ-46最佳培养时间,生物量、油脂产量、油脂含量分别为18.502 g/L、6.793 g/L、36.71%。发酵动力学分析表明:ZZ-46的底物消耗与菌体生长同步,属于与菌体生长相关型,而油脂积累则属于与菌体生长部分相关型。不饱和脂肪酸含量较高是该油的特点。这对发酵木质纤维素水解液产油脂的研究具有重要意义。Thisthesis using wheat straw and bagasse hydrolyzed with raw material preparation,on the basis of the established the possible method of detoxification,studied Trichosporon clematis ZZ -46 in hydrolysate hydrolyzed and detoxification of fermentation capacity.Using the shaking flask fermentation cultivation,the fermentation process,capacity and the fermentation kinetic parameters were analyzed.The results showed that the calcium hydroxide mixed with acticarbon was the best detoxification method.The content of furfural was the lowest,only 0.0322 μg/mL.Trichosporon dermatis ZZ -46 used detoxified lignocellulose hydrolysate to produce high oil production.The results showed that the 120 h as the best training time,and the coefficient of biomass,oil production,oil content were 18.502 g/L,6.793 g/L and 36.71%respectively.Fermentation kinetics analysis showed that the substrate consumption growth of Trichosporon dermatis ZZ-46was at the same pace with bacteria,belonging to the type was related to the growth of the bacteria,and the oil accumulation type belonged to the part related to the growth of the bacteria.The character of oil is the high content of unsaturated fatty acid.The study of lignocellulose hydrolysis fermentation to produce oil is of great significance.
