泻肝补肾汤对实验性前列腺增生大鼠前列腺重量与上皮细胞高度及组织细胞核增殖抗原表达影响

摘　　要：目的:分析泻肝补肾汤对良性前列腺增生(BPH)大鼠前列腺重量与上皮细胞高度及组织细胞核增殖抗原表达影响。方法:选取由本院实验动物中心提供的SPF级Wistar雄性大鼠30只,随机数字表法将大鼠分成三组,观察组(10只)、模型组(10只)和正常组(10只),模型组与观察组大鼠制备BPH模型,成功造模后观察组大鼠每天灌胃1ml/100g泻肝补肾汤,模型组和正常组灌胃等剂量生理盐水,连续灌胃30天,末次给药后检测各组大鼠体重、前列腺湿重及前列腺指数状况,腺体上皮细胞高度在光镜下检测,PCNA表达采用免疫组化检测。结果:模型组大鼠前列腺重量及前列腺指数较正常组显著升高,观察组大鼠前列腺重量及前列腺指数较模型组显著降低,差异均有统计学意义(P<0. 05);模型组大鼠上皮细胞高度及PCNA阳性率较正常组显著升高,观察组大鼠上皮细胞高度及PCNA阳性率较模型组显著降低,差异均有统计学意义(P<0. 05)。结论:泻肝补肾汤可降低BPH大鼠前列腺重量与上皮细胞高度,调节组织内PCNA表达对前列腺增生起抑制作用。Objective:To analyze the effect of Xiegan Bushen Decoction on prostate weight,epithelial cell height,and expression of proliferating cell nuclear antigen in tissue of benign prostatic hyperplasia (BPH)rats.Methods-Thirty male Wistar rats of SPF grade were provided by our laboratory animal center.The rats were divided into three groups by random number table method.The observation group (10),the model group (10)and the normal group,10 rats in the model group and the observation group were used to prepare the BPH model.After the successful modeling,the rats in the observation group were given 1ml/ 100g Xiebu Bushen Decoction on a daily basis,and the model group and the normal group were given an equal dose of normal sa-line by gavage for 30days.After the last administration,the body weight,prostate wet weight,and prostate index were measured.The height of glandular epithelial cells was detected under light microscope.The expression of PCNA was detected by immunohistochemistry.Results:Prostate weight and prostate index were significantly higher in the model group than in the normal group.The prostate weight and prostate index were significantly lower in the observation group than in the model group (P <0. 05),the model group had rat epithelium.The height of cells and the positive rate of PCNA were significantly higher than those in the normal group.The epithelial cell height and PCNA positive rate in the observation group were significantly lower than those in the model group,and the differences were statistically significant (P <0.05).Conclusion:Xiegan Bushen Decoction can reduce the weight of prostate and the height of epithelial cells in BPH rats,and regulate the expression of PCNA in the tissues of rats with inhibition of benign prostatic hyperplasia.
