大黄牡丹汤对重症急性胰腺炎大鼠肝肾功能及肝肾病理影响

摘　　要：目的:分析大黄牡丹汤对重症急性胰腺炎(SAP)大鼠肝肾功能及肝肾病理影响。方法:选取SPF级Wistar雄性大鼠30只,随机数字表法将大鼠分成三组,正常组(10只)、对照组(10只)及观察组(10只),观察组及对照组大鼠制备SAP模型,成功造模30分钟后观察组大鼠给予40g/kg大黄牡丹汤灌胃,对照组和正常组大鼠给予等剂量生理盐水灌胃;给药12小时后大鼠处死,心脏穿刺取血,ELISA法检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6 (IL-6)、白细胞介素-10 (IL-10)、白细胞介素-1 (IL-1)、脂肪酶及淀粉酶含量,全自动生化分析检测血清总胆红素(TBil)、肌酐(Cr)、尿素氮(BUN)、天冬氨酸转氨酶(AST)及丙氨酸转氨酶(ALT)含量,免疫组化法检测肝脏与胰腺组织内NF-κBP56蛋白的表达状况。结果:观察组和对照组大鼠血清TNF-α、IL-6、IL-10、IL-1、脂肪酶及淀粉酶含量较正常组显著升高,观察组升高幅度低于对照组,差异有统计学意义(P<0. 05);观察组和对照组大鼠血清Cr、BUN、ALT、TBil及AST含量较正常组显著升高,观察组升高幅度低于对照组,差异均有统计学意义(P<0. 05);观察组和对照组大鼠肝脏、胰腺组织内NF-κBP56阳性率较正常组显著升高,观察组升高幅度低于对照组,差异均有统计学意义(P<0. 05)。结论:大黄牡丹汤可显著降低SAP大鼠血清内炎性因子含量,改善其肝脏和胰腺病变组织,对肝肾起到保护作用。Objective:To analyze the effect of Dahuang Mudan Decoction on hepatic and renal function,pathological changes of liver and kidney in severe acute pancreatitis (SAP)rats.Methods :Thirty Wistar male SPF rats were selected and randomly divided into three groups:normal group (10),controlled group (10),and observation group (10).The observation group and the controlled group were large.Rats were prepared SAP model.30minutes after successful modeling,rats in the observation group were given 40g/kg Dahuang Mudan Decoction orally,rats in the controlled group and the normal group were given intragastric administration of equal doses of normal saline,and rats were sacrificed 12hours after administration.Blood was collected by puncture and serum TNF-α,IL-6,IL-10,IL-1 were detected by ELISA.Lipase and amylase content,automatic biochemical analysis of serum total bllirubin (TBil),ereafinine (Cr),urea nitrogen (BUN),aspartate aminotransferase (AST)and alanine aminotransferase (ALT).The contents of NF-B P56 protein in liver and pancreas were detected by immunohistochemistry. Results:The serum levels of TNF-α,IL-6,IL-10,IL-1,lipase and amylase in the observation group and the controlled group were significantly higher than those in the normal group.The increase in the observation group was lower than that in the controlled group.Statistically significant (P <0.05);Serum,BUN,ALT,TBil,and AST levels in the observation and the controlled groups were significantly higher than those in the normal group.The increase in the observation group was lower than that in the controlled group.The difference was significant (P <0.05).The positive rate of NF-B P56in liver and pancreatic tissue of the observation group and the controlled group was significantly higher than that of the normal group ;the increase rate of the observation group was lower than that of the controlled group (P <0.05).Conclusion :Dahuang Mudan Decoction can significantly reduce the content of inflammatory factors in serum of SAP rats,improve the liver and pancreatic le
