摘 要:为明确柞蚕肠道细菌的菌群结构和多样性,利用Illumina MiSeq技术检测细菌16S rRNA基因序列的方法,比较分析柞蚕5龄中期幼虫中肠和中肠内容物的细菌类群。在柞蚕5龄幼虫中肠共发现了10个门、18个纲、38个目、55个科、68个属的细菌,而在中肠内容物中仅检测到5个门、9个纲、13个目、16个科、20个属的细菌。在门的水平上,中肠的优势菌群(丰度大于1%)为蓝细菌门Cyanobacteria (86. 22%)、厚壁菌门Firmicutes(6. 22%)、变形菌门Proteobacteria (5. 55%)和放线菌门Actinobacteria (1. 49%);中肠内容物的优势菌来自蓝细菌门Cyanobacteria (93. 86%)和厚壁菌门Firmicutes (5. 18%)。在属的水平,柞蚕中肠的优势菌属为蓝细菌norankc<sub>Cyanobacteria(86. 22%)、Tyzzerella3(6. 13%)、青枯菌属Ralstonia(3. 78%)和红球菌属Rhodococcus(1. 25%);中肠内容物的优势菌属为蓝细菌norankc<sub>Cyanobacteria(93. 86%)和Tyzzerella3(5. 08%)。水小杆菌属Aquabacterium等13个属在中肠的丰度是中肠内容物的4. 92130. 32倍,而51个属在中肠内容物中没有检测到。中肠菌群的Chao指数和Ace指数均明显高于中肠内容物。结果表明,柞蚕中肠和中肠内容物的菌群结构在组分和丰度上均存在明显差异,总体上中肠内容物的菌群远不如中肠的菌群丰富。To understand bacterial community structure and diversity in the intestine of Chinese oak silkworm,Antheraea pernyi,bacteria communities of the midgut,as well as the midgut contents,in middle stage of the 5 th instar larvae were investigated based on 16S rRNA gene sequences through the Illumina MiSeq technology.We acquired bacteria from 10 phyla,18 classes,38 orders,55 families and 68 genera in the midgut,but only 5 phyla,9 classes,13 orders,16 families and 20 genera in.the midgut contents.At the phylum level,the dominant bacterial group (abundance >1%)in midgut was Cynobacteria (86.22%),Firmicutes (6.22%),Proteobacteria (5.55%)and Actinobacteria (1.49%), whereas that in midgut contents was Cyanobacteria (93.86%)and Firmicutes (5.18%).At the genus level,the pre-dominant bacteria in the midgut were from norank c Cyanobacteria (86.22%),Tyzzerella_3(6.13%),Ralstonia (3.78%)and Rhodococcus (1.25%),whereas those in the midgut contents were from norank_c__Cyanobacteria (93.86%)and Tyzzerella_3(5.08%).The abundance of 13 genera including Aquabacterium in the midgut was 4.92to 130.32times higher than those in the midgut contents,and 51genera that occurred in the midgut were not detected in the midgut contents.The indices of Chao and Ace of bacteria in the midgut were obviously higher than those in the midgut contents.The above results presented here showed an obvious difference in the composition and abundance of intestinal bacteria between the midgut and its contents.In general,the bacterial composition and diversity of the midgut contents is far less than that of the midgut.
