摘 要:目的研究不同品系小鼠对葡萄糖耐量的变化规律,并通过曲线拟合方法进行对比分析。方法以4种不同品系雄性小鼠(体质量23~25 g)为研究对象,并按小鼠品系分为4组:C57组、BALB/c组、CD1组和KM组(昆明小鼠)。小鼠禁食12 h后,血糖仪检测各组小鼠初始血糖值。随后立即灌胃给予0. 2 ml 15%葡萄糖溶液,并分别在灌胃后5、10、15、30、60和120 min检测血糖值。结果重复测量资料方差分析显示,不同品系小鼠灌胃后不同时间点血糖变化值(相对0 min)差异具有统计学意义(F(3,36)=30. 003,P=0. 000);在灌胃约15 min,各品系小鼠血糖值达到峰值,约120 min回归正常水平;各品系小鼠葡萄糖耐量变化规律一致,均符合Giddings曲线拟合模型(R2≥0. 95)。结论上述4种品系小鼠虽初始血糖水平有所差异,但在葡萄糖耐量变化规律方面有较好的一致性,符合Giddings曲线变化特征,为后续定量研究小鼠的糖耐量规律及糖代谢异常相关研究提供了参考。Objective To explore the blood glucose tolerance in different strains of mice,and to establish a mathematical model of glucose metabolism by curve fitting analysis.Methods Four different strains of mice (relative body mass 23-25g)were used as subjects.The mice were divided into four groups :C57group,BALB/c group,CD1group and KM group according to their strains.After 12h of fasting,the initial blood glucose of each group of mice was monitored by the Roche Accu-Chek Softclix glucometer.Then,each animal was administered with 0.2ml of 15%glucose solution by gavage immediately.The blood glucose levels in animals were measured at 5,10,15,30,60and 120min.Results There was significant difference in the changes of the blood glucose levels between different strains of mice according to the repeated measurement data (F(3.36)=30.003,P =0.000).The blood glucose level of each strain of mice peaked at about 15min after glucose administration,and returned to normal at about 120min.According to the Giddings model by the curve fitting analysis,the glucose metabolism of each strain of mice was consistent (R^2≥0.95).Conclusion The glucose metabolism of each strain has a good consistency in the blood glucose peak as well as the glucose metabolism time and glucose metabolism curve model.In addition,the Giddings curve fitting model is consistent with the glucose metabolism of mice.This finding can contribute to further study of the glucose metabolism and abnormal glucose metabolism in diabetes related studies.
