摘  要:目的:采用高效液相色谱法测定不同产地及不同采收期凌霄花中麦角甾苷的含量,比较不同产地、不同采收期凌霄花的质量差异。方法:采用Ultimate XB-C18(250mm×4.6mm,5μm)色谱柱,柱温25℃,以乙腈-0.1%冰醋酸水溶液为流动相,梯度洗脱,流速1.0mL·min^(-1),检测波长334nm。结果:麦角甾苷浓度在25.75-206.0μg/mL范围内与峰面积呈良好的线性关系,r=0.9999。平均回收率为104.3%,(RSD=0.9%,n=6)。不同产地的凌霄花中,麦角甾苷以江苏连云港采收的含量最高,山东产的凌霄花中麦角甾苷含量在叶部的含量明显高于花部与茎部。结论:本方法操作简单、快速、重复性好,可用于测定凌霄花中麦角甾苷的含量。Objective:This research aims to establish a HPLC method for the determination of acteoside in the different parts of Campsis grandiflora from different habitats.Methods:The chromatographic separation was performed on Ultimate XB C18 column(4. 6× 250 mm,5μm)with gradient elution consisting of acetonitrile and water(containing 0. 1% acetic acid)at a flow rate 1. 0 mL/min. The UV detection wavelength was 334 nm and the column temperature was set at 25 ℃.Results:The linear ranges of acteoside was 25. 75-206. 0μg/mL,r=0. 9999. The average recovery of acteoside was 104. 3%(RSD=0. 9%,n=6). The content of acteoside in the sample collected from Lianyungang city,Jiangsu province is the highest;The content of acteoside in the leaves of Campsis grandiflora from Shandong province is obviously higher than that of in the flowers and the stems.Conclusion:The validated HPLC method has the advantages of precision,reproducibility and reliability,allowing the determination of acteoside in Campsis grandiflora.
