摘  要:目的研究β-榄香烯对结肠癌细胞系SW480的放疗增敏作用,并分析其对磷脂酰肌醇3-激酶/丝苏氨酸蛋白激酶(PI3K/Akt)信号通路的影响。方法将细胞分为6组:空白对照组、辐照组(进行4 Gy辐照)、药物组(10μmol·L^(-1)β-榄香烯)、抑制剂组(50μmol·L^(-1)PI3K抑制剂,LY294002)、联合1组(4 Gy辐照+10μmol·L^(-1)β-榄香烯)、联合2组(4 Gy辐照+50μmol·L^(-1)LY294002)。用噻唑蓝法检测细胞增殖抑制率,用流式细胞术检测细胞凋亡和细胞周期分布,用免疫印迹法测定相关蛋白表达。结果抑制剂组、药物组、辐照组、联合2组和联合1组细胞凋亡率分别为(20. 39±1. 34)%,(21. 62±1. 23)%,(8. 94±1. 16)%,(31. 76±4. 22)%和(32. 83±4. 17)%;这5组的G0/G1期DNA含量分别为52. 68±4. 13,54. 36±4. 68,49. 20±4. 77,74. 63±6. 29和74. 92±6. 34;这5组的细胞集落形成数目分别为111. 19±15. 33,108. 74±15. 28,116. 67±14. 39,45. 32±9. 06和40. 08±8. 43;这5组的细胞内p Akt蛋白表达水平分别为0. 86±0. 04,0. 85±0. 04,0. 87±0. 05,0. 42±0. 03和0. 41±0. 03。上述指标,联合2组、联合1组与抑制剂组、药物组和辐照组相比,差异均有统计学意义(均P <0. 01)。结论β-榄香烯能够通过抑制PI3K/Akt信号通路激活,抑制周期相关蛋白表达引起G2/M期阻滞以抑制细胞增殖,同时促进细胞凋亡相关蛋白表达诱导细胞凋亡,从而提高SW480细胞的放射敏感性。Objective To investigate the radiosensitizing effect of β - elemene on human colon carcinoma cell line SW480 and its effect on the signal pathway of phosphatidylinositol 3-kinase(PI3K) / serine threonine protein kinase (Akt) .Methods The cells were divided into 6 groups: blank control group,irradiation group (4 Gy irradiation) ,drug group (10 μmol·L^-1 β-elemene) ,inhibitor group (50 μmol·L^-1 PI3K inhibitor,LY294002) ,combined-1 group (added 10 mmol·L^-1 β-elemene immediately after 4 Gy irradiation) ,combined-2 group (added 50 μmol·L^-1 LY294002 immediately after irradiation of 4 Gy) . The inhibitory rate of cell proliferation was detected by MTT assay.Apoptosis and cell cycle distribution were detected by flow cytometry,and protein expression was analyzed by Western blot.Results The apoptotic rates in inhibitor group,drug group,irradiation group,combined-2 group and combined-1 groups were(20.39±1.34) %,(21.62±1.23) %, (8.94±1.16) %,(31.76±4.22) %,(32.83±4.17) % .The DNA contents in G0 /G1 phase in the 5groups were 52.68 ±4.13,54.36 ±4.68,49.20±4.77,74.63±6.29,74.92±6.34.The colony formation rates in the 5 groups were 111.19±15.33, 108.74±15.28,116.67±14.39,45.32±9.06,40.08±8.43.The expression levels of pAkt protein among the 5 groups were 0.86±0.04,0.85±0.04,0.87±0.05,0.42±0.03,0.41±0.03.Compared between combined-2 group,combined-1 group,inhibitor group,drug group and irradiation group,the differences of the factors were significant (all P<0.01) .Conclusion β-elemene can inhibit cell proliferation by inhibiting the PI3K/Akt signaling pathway and G2 /M arrest in cell cycle,and promote the apoptosis-related protein expression,leading to improve the radiosensitivity of SW480 cells.
