摘 要:目的 评价外源性消退素D2对大鼠非压迫性腰椎间盘突出症根性神经痛的影响.方法 清洁级健康雄性SD大鼠36只,8周龄,体重230~ 270 g,采用随机数字表法分为3组(n=12):假手术组(S组)、非压迫性腰椎间盘突出症根性神经痛组(P组)和外源性消退素D2组(R组).鞘内置管后3d,S组仅暴露相应手术部位;P组和R组取自体髓核组织覆盖于右侧L5背根神经节制作非压迫性腰椎间盘突出症根性神经痛模型.造模后3d内每天鞘内注射相应药物,P组注射10μl PBS溶液,R组注射10μl 1 ng/μl消退素D2溶液,给药后3组均以10μl生理盐水冲管.造模前1d和造模后1~7d每天测定术侧机械缩足反应阈(MWT).造模后第7天取术侧腰膨大段脊髓背角,用Western blot法测定胶质纤维酸性蛋白(GFAP)的表达,免疫荧光双标法检测G蛋白偶联受体18与GFAP共表达情况.结果 与S组比较,P组造模后1~7 d MWT降低,GFAP表达上调(P<0.05);与P组比较,R组造模后3~7 d MWT升高,GFAP表达下调(P<0.05).G蛋白偶联受体18与GFAP存在共表达.结论 外源性消退素D2可减轻大鼠非压迫性腰椎间盘突出症根性神经痛,其机制与抑制脊髓背角星形胶质细胞活化有关.Objective To evaluate the effect of exogenous resolvin D2on radieular pain in a rat model of non-compressive lumbar intervertebral disc herniation.Methods Thirty-six clean-grade healthy male Sprague-Dawley rats,aged 8weeks,weighing 230-270g,were divided into 3groups (n=12each) using a random number table method :sham operation group (group S),radieular pain induced by noncompressive lumbar intervertebral disc herniation group (group P)and exogenous resolvin D2group (group R).The right L5dorsal root ganglions were covered by autologous nucleus pulposus tissues to establish the model of non-compressive lumbar disc herniation in P and R groups.The corresponding surgical site was on- ly exposed in group S.The corresponding drugs were intratheeally injected within 3days after establishing the model,phosphate buffer solution 10μlwas injected in group P,1ng/μl resolvin D2solution 10μl was injected in group R,and normal saline 10μlwas given for pipe washing after administration in the three groups.The mechanical paw withdrawal threshold (MWT)was measured on 1day before establishing the model and 1-7days after establishing the model.The spinal dorsal horns of lumbar enlargement segments were removed on day 7after establishing the model for determination of the expression of glial fibrillary acid-ic protein (GFAP)(by Western blot)and co-expression of G-protein-coupled receptor 18with GFAP (by double-label immunofluorescence).Results Compared with group S,the MWT was significantly decreased at 1-7days after establishing the model,and the expression of GFAP was up-regulated in group P (P<0.05).Compared with group P ,'the MWT was significantly increased at 3-7days after establishing the model,and the expression of GFAP was down-regulated in group R (P<0.05).G-protein-coupled receptor 18was co-expressed with GFAP.Conclusion Exogenous resolvin D2can reduce radicular pain in a rat model of non-compressive lumbar intervertebral disc herniation,and the mechanism is related to inhibiting activation of astrocytes in the spinal d
