摘 要:目的研究神经节苷脂(GM1)对大鼠急性脊髓损伤后Caspase-3水平表达的影响,探讨GM1抑制神经细胞凋亡作用机制。方法选择75只成年健康SD大鼠,体重226~365 g,随机分为3组,按照环扎法建立急性脊髓损伤模型。根据随机数字分为假手术组25只(A组),脊髓损伤组25只(B组),GM1组25只(C组),三组根据时间点不同分为1 d、7 d和14 d三个不同亚组。术后观察大鼠一般情况,同时分别于伤后1 d、7 d、14 d采用斜板试验和BBB评分进行神经功能评估,取材观察脊髓形态变化,免疫组织化学染色法观察三组大鼠脊髓损伤部位Caspase-3的表达,TUNEL标记观察细胞凋亡水平。结果 B组和C组共死亡8只SD大鼠,每组纳入20只大鼠进行观察。各时间点A组斜板试验及BBB评分均优于B组、C组(P<0.05),术后各时间点C组斜板试验及BBB评分均明显优于B组(P<0.05)。组织学观察显示,A组各时间点脊髓组织结构完整,术后1 d、7 d及14 d脊髓水肿坏死程度C组轻于B组。各时间点B组、C组Caspase-3阳性细胞数及TUNEL阳性细胞数均明显多于A组(P<0.05),B组Caspase-3阳性细胞数及TUNEL阳性细胞数多于C组,差异有统计学意义(P<0.05)。结论 GM1可能通过下调Caspase-3蛋白表达抑制神经细胞凋亡治疗急性脊髓损伤。Objective To study the effect of ganglioside(GM1) on the expression of Caspase-3 in rats after acute spinal cord injury,and to explore the mechanism of anti apoptosis effect on neural cell.Methods Seventy-five adult healthy SD rats weighing from 226 to 365 g were randomly divided into three groups:sham operation group(group A,n = 25),spinal cord injury group(group B,n = 25),and GM1 group(group 3,n =25).The acute spinal cord injury model was established according to the cerclage method.The three groups of rats were divided into three different subgroups according to time points(1 d,7 d and 14 d).The general condition of the rats was observed.The sloping plate test and BBB scores were used to evaluate the neurological function at1 st,7 th and 14 th day after injury respectively.The histological changes of the spinal cord were observed.Expression of Caspase-3 at the spinal cord injury sites of the three groups were observed by immunohistochemical staining,and TUNEL method was used to measure the level of cell apoptosis.Results A total of 8 SD rats were sacrificed in group B and group C,and 20 rats in each group.The slope plate test and BBB score of group A were better than those of groups B and C at each time point(P<0.05).The slope plate test and BBB scores of group C were significantly better than those of group B at each time point(P<0.05).Histological observation showed that the spinal cord tissue was integrated at each time point in group A.The degree of spinal edema and necrosis at group C was lower than that of group B in postoperative time points:1 st day,7 th day and 14 th day.The number of Caspase-3 positive cell and TUNEL positive cell in group B and C was significantly higher than that in group A at each time point(P<0.05).The number of Caspase-3 positive cell and TUNEL positive cell in group B was more than that in group C,and there was a statistically significant difference(P < 0.05).Conclusions GM1 may inhibit neuronal apoptosis by down-regulating Caspase-3 protein expression in the treatment of acute
