益气固本胶囊对哮喘小鼠CD4^+CD25^+Foxp3^+Treg细胞数量及IL-33、TNF-α表达水平的影响

摘　　要：目的观察不同剂量益气固本胶囊对哮喘小鼠CD4^+CD25^+Foxp3^+Treg细胞数量及白细胞介素-33(IL-33)、肿瘤坏死因子-α(TNF-α)表达水平的影响。方法将32只雄性SPF级Balb/c小鼠随机分成正常对照组、模型组、益气固本胶囊低剂量组和益气固本胶囊高剂量组,每组8只。除正常对照组外将其他组中小鼠应用卵白蛋白和氢氧化铝混合液腹腔致敏和激发复制哮喘模型,于第21天开始,在每次激发前1h分别对正常对照组和模型组小鼠给予生理盐水灌胃,用药各组小鼠给予药物灌胃,连续10d。应用ELISA双抗体夹心法测定血清中IL-33、TNF-α水平,流式细胞仪检测脾淋巴细胞中CD4^+CD25^+Foxp3^+Treg/CD4^+T比例。结果模型组小鼠血清中IL-33水平较正常对照组水平低(P<0.05),而血清中TNF-α水平较正常对照组明显增高(P<0.01)。益气固本胶囊低剂量组和高剂量组血清中的IL-33水平高于模型组(P<0.05),血清中的TNF-α水平低于模型组(P<0.05)。模型组脾淋巴细胞中的CD4^+CD25^+Foxp3^+Treg/CD4^+T细胞比例与正常组比较明显减少,而益气固本胶囊高剂量组明显高于模型组(P<0.05)。结论CD4^+CD25^+Foxp3^+Treg参与哮喘的发病过程,益气固本胶囊可能通过上调CD4^+CD25^+Foxp3^+Treg细胞数量及降低TNF-α水平及升高IL-33水平,调节免疫失衡,减轻哮喘炎症。Objective To observe the effects of different doses of Yiqiguben capsule on the number of CD4^+ CD25^+ Foxp3^+ Treg cells and expression levels of interleukin-33 ( IL-33) and tumor necrosis factor ( TNF-α) in mice with asthma.Methods Thirty-two male SPF grade Balb /c mice were randomly divided into normal control group,model group,Yiqiguben low-dose group,and Yiqiguben high-dose group,8 rats in each group.Apart from normal control group,the mice in the other groups were treated with combination of ovalbumin and aluminum hydroxide for peritoneal sensitization to challenge and duplicate asthma model.From the 21st day,the mice in normal control group and model group were given intragastric administration of normal saline,and the mice in Yiqiguben low-dose group and Yiqiguben high-dose group were given intragastric administration of Yiqiguben at one hour before each challenge for consecutive 10 days.ELISA double-antibody sandwich method was used to detect the levels of IL-33 and TNF-α in serum.Flow cytometry was used to detect the ratio of CD4^+CD25^+Foxp3^+Treg /CD4^+T cells in splenic lymphocytes.Results The level of serum IL-33 in model group was statistically significantly lower than that in normal control group ( P<0.05) ,while the level of serum TNF-α in model group was statistically significantly higher than that in normal control group ( P<0.01) .The levels of serum IL-33 in Yiqiguben low-dose group and Yiqiguben high-dose group were statistically significantly higher than that in model group ( P<0.05) ,while the levels of serum TNF-α in Yiqiguben low-dose group and Yiqiguben high-dose group were statistically significantly lower than that in model group ( P<0.05) .The proportion of CD4^+CD25^+Foxp3^+ Treg /CD4^+T cells in splenic lymphocytes in model group was statistically significantly lower than that in normal group,while the proportion of CD4^+CD25^+Foxp3^+Treg /CD4^+T cells in splenic lymphocytes in Yiqiguben high-dose group was statistically significantly higher than that in model group ( P<0.05) .
