摘 要:目的通过检测Bid在哮喘大鼠肺组织中表达的变化,探讨Bid在哮喘炎症机制中的作用。方法通过卵白蛋白致敏复制哮喘大鼠模型,分别采用免疫组化法及实时定量PCR法检测肺组织Bid mRNA和蛋白的表达。结果哮喘组Bid mRNA和蛋白的表达水平显著高于对照组(P <0. 01),地塞米松组Bid蛋白的表达水平显著低于哮喘组(P <0. 05),而地塞米松组Bid mRNA的表达量与哮喘组比较差异无统计学意义(P>0. 05),肺组织Bid mRNA与蛋白的表达水平无显著相关性(n=24,r=0. 39,P>0. 05)。结论哮喘大鼠急性发作期肺组织Bid的表达升高,可能在哮喘的炎症机制中起着促进气道炎症的作用,这种促炎的功能可以被地塞米松所抑制。Objective To investigate the potential roles of Bid in asthmatic inflammation pathogenesis by investigating the changes of Bid expressions in rat asthma lung tissue. Methods Rat asthma model were copied by ovalbumin,and the levels of Bid mRNA and protein in rat lung tissue were detected by immunohistochemisty and real-time polymerase chain reaction methods. Results Levels of Bid mRNA and protein at lung tissue in asthma group were significantly higher than those in control group( P < 0. 01). Moreover,levels of Bid protein in desamethasone group was significantly lower than that in asthma group( P < 0. 05),while it was no statistical significance on the Bid mRNA difference compared with that in desamethasone group( P > 0. 05). Furthermore,there was no statistical correlation between levels of Bid mRNA and protein( n = 24,r =0. 39,P > 0. 05). Conclusion Bid level increased in lung tissue of asthmatic rats,which may act as a key role to promote asthmatic airway inflammation,but this function can be regulated by desamethasone.
