摘 要:目的探讨新生雄性小鼠泌尿生殖系统显微三维重建的可行性,以便为泌尿生殖系统发育及畸形的研究提供更精确的方法学基础。方法对新生雄小鼠泌尿生殖系统进行整块原位固定,连续组织切片,并经图像配准,然后用计算机软件进行三维重建,并测算正常及己烯雌酚(DES)影响后新生小鼠泌尿生殖系统有关器官的形态参数。结果正常组新生小鼠泌尿生殖系统连续切片显微三维重建显示各器官形态、位置良好,实验组显示肾脏、睾丸、附睾及睾丸引带等重要器官形态、位置异常明显。这些正常及异常的器官形态和位置可经三维重建软件进行全部或部分的三维任意角度旋转显示,并可具体量化到微米级。结论新生雄小鼠泌尿生殖系统可经一定方法进行显微组织三维重建,任意角度立体显示并精确量化。该方法可为泌尿生殖系统的各种形态学研究所采用,或为其他系统器官的形态学研究所借鉴。Objective To explore the feasibility of three dimensional (3D)reconstruction of urogenital system in neonatal male mice so as to provide a more accurate methodological basis for examining urogenital development and malformation.Methods The whole urogenital system of neonatal male mice was fixed in situ and sliced in series.And serial micrographs were acquired and then 3D reconstruction was processed with special software.At the same time,the morphological parameters of urogenital organs were evaluated in normal and diethylstilbestrol (DES)-treated groups. Results 3D histological reconstruction with serial urogenital slides showed normal contour and location in normal group.While in experimental group,contour and location of vital organs were abnormal.The abnormalities could be displayed wholly or partially by 3D reconstruction software with three-dimensional rotation at any angle.And the details were further quantified to a micrometer level. Conclusions The urogenital system of neonatal male mice may be reconstructed by 3D histology.This method is applicable for various morphological studies of urogenital system or other organs.
