摘  要:目的:研究白芨水提物对人肝癌HepG2细胞增殖的影响。方法:使用水提醇沉法提取白芨粗多糖,同时保留有机小分子物质部分;采用CCK8试剂检测不同浓度的两部分提取物对HepG2细胞存活率及生长曲线的影响;倒置显微镜观察提取物对HepG2细胞形态的影响,以及soft agar法检测提取物对HepG2细胞克隆形成的影响。结果:与对照组相比,白芨水提物均能降低HepG2细胞的存活率和增殖能力,并与浓度呈正相关,其中多糖和有机小分子物质的半数抑制浓度(IC_(50))分别为65. 99μg/mL和52. 62μg/mL;此外,白芨水提物作用后改变了细胞形态,并能抑制HepG2细胞的克隆形成能力。结论:白芨水提物中的多糖和有机小分子物质均能抑制HepG2细胞的增殖作用。Objective To study the effect of the aqueous extract of Bletilla striata on proliferation of human hepatocarcinoma cell line HepG2.Methods Crude polysaccharides of Bletilla striata were extracted byaqueous extraction and alcohol precipitation,while the part of organic smallmolecule was preserved.After treatment with different concentrations of extracts,the cell viability and growth curve were detected by CCK8assay,and cell morphology was observed by inverted microscope,as well as the formation of HepG2 cell clones were detected by soft agarassay.Results Compared with control group,the HepG2cell viability and proliferation were reduced ina dose-dependent manner,and the ICs0values of polysaccharide and organic small molecule were 65.99 μg/mL and 52.62 μg/mL, respectively.In addition,the cell morphology was changed and the formation of HepG2cell clones wereinhibited after cultured with a- queous extract of Bletilla striata.Conclusion Both polysaccharide and organic small molecules in the aqueous extract of Bletilla striatacan inhibit the proliferation of HepG2 cells.
