摘  要:目的观察石斛合剂对db/db糖尿病肾病小鼠水代谢相关指标的影响。方法选用8~9周龄雄性db/db小鼠,按空腹血糖及体质量随机分成模型组、六味地黄丸组、石斛合剂低、高剂量组,db/m小鼠为正常组。除模型组和正常组灌生理盐水外,其余各组灌胃相应的药物,连续给药8周。实验期间,分别于第0周、4周、8周测定各组空腹血糖(FBG),实验结束后测定血清肌酐(Scr)、尿素氮(BUN)含量,观察肾病理变化,并用免疫荧光和Western-blot法检测AQP2、AQP3的表达。结果与模型组比较,石斛合剂低、高剂量组和六味地黄丸组干预8周后,都可以显著降低db/db小鼠的空腹血糖,并降低血清中Scr和BUN(P<0.05或P<0.01);模型组AQP2、AQP3的蛋白表达较正常组明显升高(P<0.01),石斛合剂高剂量组可下调AQP2、AQP3的异常高表达(P<0.05或P<0.01)。结论石斛合剂能有效改善db/db糖尿病小鼠的高血糖和肾功能,其对糖尿病肾病的防治作用可能与调节水通道蛋白的表达相关。Objective:To investigate Shihu Mixture regulation of related indicators on water metabolism in db/db diabetic nephropathy mice.Methods:8 to 9-week-old male db/db mice were randomly divided into model group,Linwei Dihuang Pill positive treatment group,Shihu Mixture low-dose group and high-dose group according to fasting blood glucose and body weight,and with db/m mice as normal control group.Normal control group and model group were given normal saline,while other groups were given relevant medicines intragastrically once a day for consecutive 8 weeks.During the experimental period,the fasting blood glucose (FBG)test was performed periodically.The semm creatinine (Scr)and urea nitrogen (BUN)levels in blood were measured.The kidneys were paraffin-embedded and examined for pathological changes.Immunofluorescence and Western-blot were used to detect AQP2 and AQP3 expression.Results:Shihu Mixture could reduce fasting blood glucose,improve renal function significantly in db/db mice,and regulate renal tubular AQP2 and AQP3 protein expression.Conclusion:Shihu Mixture can recover hyperglycemia and renal function in db/db diabetic mice effectively.The Shihu mixture prevention and treatment of diabetic nephropathy is related to the regulation of water metabolism.
