肾安冲剂对阿霉素肾病综合征大鼠Ⅳ型胶原和纤维连接蛋白的影响

摘　　要：目的观察肾安冲剂对阿霉素肾病综合征大鼠Ⅳ型胶原(Ⅳ-C)和纤维连接蛋白(FN)表达的影响。方法肾病综合征大鼠模型采用阿霉素(ADR)尾静脉注射造模。SD雄性大鼠72只随机分为6组:肾安冲剂大剂量组、中剂量组、小剂量组,泼尼松组,正常对照组和模型组,每组各12只。造模后开始给药,正常对照组、模型组予生理盐水,泼尼松组灌服泼尼松6.45 mg/(kg·d),肾安冲剂大剂量组、中剂量组和小剂量组分别灌服肾安冲剂10.32 g/(kg·d)、5.16 g/(kg·d)、2.58 g/(kg·d)。8周后,取大鼠肾脏做常规石蜡切片,免疫组化观察其Ⅳ-C和FN表达的程度。结果模型组肾组织Ⅳ-C和FN表达明显增强,肾安冲剂小剂量组、中剂量组、大剂量组和泼尼松组均可降低Ⅳ-C和FN表达(P<0.01),以肾安冲剂大剂量组疗效最好(P<0.01)。结论肾安冲剂可降低阿霉素肾病综合征大鼠肾脏Ⅳ-C和FN表达,减少其对肾脏的损害,延缓肾间质纤维化和肾小球硬化的进程。Objective:To investigate the effect of Shen-An Chongji (SACJ)on collagen type IV (IV-C)and fibronectin (FN)of kidney in rats with adriamycin nephropathy.Methods:Seventy-two male SD rats were randomly divided into fiormal control group, mode group,prednisone group,SACJ low dose group (SACJ-L),moderate dose (SACJ-M)group and high dose (SACJ-H)group, with 12 rat in each group.Rats in the latter 5 groups were made into nephrotic syndrome model by tail intravenous injection with adriamycin.Drugs administration of started after the models made,and lasted for 8 weeks.The IV-C and FN expressions in rat kidney were determined by immunohistoehemical assay.Results:Expressions of IV-C and FN in kidney were higher ha the mode group than normal control group,which were lowered in group SACJ-L,SACJ-M,SACJ-H and the prednisone (P<0.01),and the optimal effect was shown in group SACJ-H (P<0.01).Conclusion:SACJ could reduce expression of IV-C and FN in rats with adriamycin nephropathy,relieve the lesion of kidney owing to the pathologic agents,slow the processes of glomerulosclerosis and renal interstitial fibrosis. and the effect of SACJ was suoerior to that of use orednisone.
