摘 要:目的探讨针刺对老龄大鼠全脑缺血再灌注时海马胆碱能抗炎通路的影响。方法清洁级老龄健康雄性SD大鼠96只,17~22月龄,体重440~580g,采用随机数字表法分为3组(n=32):假手术组(S组)、全脑缺血再灌注组(I/R组)和针刺组(AP组)。I/R组和AP组采用四动脉阻断法制备大鼠全脑缺血再灌注损伤模型,AP组于缺血前针刺百会和风池穴,连续针刺14d。分别于再灌注1、3、5和7d时随机处死4只大鼠,取脑组织,采用TUNEL法测定海马CA1区神经元凋亡率。分别于再灌注1、3、5和7d时随机处死4只大鼠,取脑组织,采用Western blot法检测海马CA1区α7烟碱型乙酰胆碱受体(α7nAChR)、胆碱乙酰基转移酶(ChAT)、TNF-α和IL-1β表达。结果与S组比较,I/R组和AP组再灌注各时点海马神经元凋亡率升高,α7nAChR和ChAT、TNF-α和IL-1β的表达上调(P<0.05);与I/R组比较,AP组再灌注各时点海马神经元凋亡率降低,α7nAChR和ChAT的表达上调,TNF—α和IL-1β的表达下调(P<0.05)。结论针刺减轻老龄大鼠全脑缺血再灌注损伤的机制可能与激活海马胆碱能抗炎通路有关。Objective To investigate the effect of acupuncture on cholinergic anti-inflammatory pathway in the hippocampus of aged rats during global cerebral ischemia-reperfusion (I/R).Methods Ninety-six clean-grade healthy male Sprague-Dawley rats,aged 17-22 months,weighing 440-580g,were divided into 3 groups (n =32each)using a random number table method:sham operation group (group S),global cerebral I/R group (group I/R)and acupuncture group (group AP).Global cerebral I/R was induced by 4-vessel oeelusion method described by Pulsinelli in group I/R and group AP.Baihui and Fengchi were stimulated for 14consecutive days before ischemia in group AP.Four rats were sacrificed at 1,3, 5 and 7 days of reperfusion,and brains were removed for determination of neuronal apoptosis by TUNEL. Four rats were sacrificed at 1,3,5 and 7 days of reperfusion,and brains were removed for determination of the expression of α7 nicotinic acetylcholine receptor (α7 nAChR),choline acetyhransferase .(CHAT), tumor necrosis factor-α (TNF-α)and interleukin-1β(IL-1β)in the hippocampal CA1 region by Western blot.The apoptosis rate was calculated.Results Compared with group S,the apoptosis rate of hippocampal neurons was significantly inereased,and the expression of α7nAChR,CHAT,TNF-α and IL-1β was up-regulated at each time point of reperfusion in I/R and AP groups (P<0.05).Compared with group I/R, the apoptosis rate of hippocampal neurons was significantly decreased,the expression of α7nAChR and ChAT was up-regulated,and the expression of TNF-α and IL-1β was down-regulated at each time point of reperfusion in group AP (P<0.05).Conclusion The mechanism by which acupuncture mitigates global cerebral I/R injury may be related to activating cholinergie anti-inflammatory pathway in the hippocampus of aged rats.
