摘 要:目的 探讨右美托咪定预先给药对大鼠炎症性内脏痛的影响.方法 健康雄性Wistar大鼠40只,体重200~300 g,6~8周龄,采用随机数字表法分为5组(n=8):对照组(C组)、内脏痛组(VP组)、右美托咪定1μg∕kg组(Dex1组)、右美托咪定5μg∕kg组(Dex2组)和右美托咪定10μg∕kg组(Dex3组).VP组、Dex1组、Dex2组和Dex3组采用腹腔注射0.9%乙酸10 ml∕kg制备炎症性内脏痛模型,C组腹腔注射等容量生理盐水.腹腔给药前15 min时,Dex1组、Dex2组和Dex3组分别经尾静脉注射右美托咪定1、5和10μg∕kg,C组和VP组尾静脉注射等容量生理盐水.观察造模60 min内大鼠行为学变化,计算内脏痛指数(VPI).造模后180 min时心脏采血样,测定血清TNF-α浓度,然后处死大鼠,取结肠组织,光镜下观察病理学结果.结果 与C组比较,VP组和Dex1-2组VPI和血清TNF-α浓度升高,Dex3组血清TNF-α浓度升高(P<0.05),VPI差异无统计学意义(P>0.05);与VP组比较,Dex1-3组VPI和血清TNF-α浓度降低(P<0.05),结肠组织病理学损伤减轻;与Dex1组比较,Dex2-3组VPI和血清TNF-α浓度降低(P<0.01),结肠组织病理学损伤减轻;与Dex2组比较,Dex3组VPI和血清TNF-α浓度降低(P<0.01),结肠组织病理学损伤减轻.结论 右美托咪定预先给药可减轻大鼠炎症性内脏痛.Objective To investigate the effect of dexmedetomidine pretreatment on inflammatory visceral pain in rats. Methods Forty healthy male Wistar rats, weighing 200-300 g, aged 6-8 weeks, were divided into 5 groups ( n=8 each) using a random number table method: control group ( group C) , visceral pain group ( group VP ) , dexmedetomidine 1 μg∕kg group ( group Dex1 ) , dexmedetomidine 5μg∕kg group ( group Dex2) and dexmedetomidine 10μg∕kg group ( group Dex3) . The model of inflammato-ry visceral pain was established by intraperitoneally injecting 0. 9% acetic acid 10 ml∕kg in VP, Dex1, Dex2 and Dex3 groups, and the equal volume of normal saline was given instead in group C. At 15 min be-fore intraperitoneal injection, dexmedetomidine 1, 5 and 10μg∕kg were injected via the tail vein in Dex1, Dex2 and Dex3 groups, respectively, and the equal volume of normal saline was given instead in C and VP groups. Behavioral changes of rats were observed within 60 min after the model was established, and viscer-al pain index ( VPI) was calculated. Blood samples were collected from the hearts at 180 min after establis-hing the model for determination of tumor necrosis factor-alpha ( TNF-α) concentrations in serum. The ani-mals were then sacrificed, and colons were obtained for examination of pathological changes with a light mi-croscope. Results Compared with group C, the VPI and serum TNF-α concentrations were significantly increased in VP and Dex1-2 groups, and the serum TNF-αconcentrations were significantly increased ( P<0. 05), and no significant change was found in VPI in group Dex3 (P>0. 05). Compared with group VP, the VPI and serum TNF-α concentrations were significantly decreased (P<0. 05), and the pathological changes of colon tissues were significantly attenuated in group Dex1-3. Compared with group Dex1, the VPI and serum TNF-α concentrations were significantly decreased ( P<0. 01) , and the pathological changes of colon tissues were significantly attenuated in Dex2-3 groups.
