摘 要:目的探索酸化甘油溶血试验在红细胞储存损伤研究中应用的可能性。方法随机采集无偿献血者全血标本20份,4℃储存5d。分别于储存d1,d3和d5采用AGLT和EOFT来检测渗透脆性。AGLT计算50%未溶血率对应的时间,EOFT计算50%溶血率对应的NaCl浓度。比较2种试验方法在检测红细胞渗透脆性上的差异。结果AGLT50%未溶血率所对应的时间从(34.60±1.40)s缩短到(28.71+±2.50)s(P<0.05),EOFT50%溶血率所对应的NaCl浓度从(4.36±0.21)g/L增加到(4.89±0.26)g/L(P<0.05);AGLT在性别之间无显著差异(P>0.05),EOFT在性别之间也无显著差异(P>0.05)。结论AGLT计算结果精确,物料耗费少,操作耗时短,是一种可以用于红细胞储存损伤研究的检测方法。Objective To explore the possibility of applying the acidified glycerol hemolysis test in the lesion study of red blood cells during storage. Methods Samples of 20 blood donors were randomly collected and stored at 4 ℃ for 5 days.Samples were tested on 1,3,5 days respectively by AGLT and EOFT. Time of the 50% non-hemolysis rate were calculated in AGLT and Na Cl concentration of 50% hemolysis rate were calculated in EOFT. Comparison of the differences between the two methods in detecting osmotic fragility of red blood cells was made. Results In AGLT,the time of 50% non-hemolysis rate shortened with the extension of storage time( P<0. 05). In EOFT,the Na Cl of 50% hemolysis rate increased with extended storage time( P<0. 05). AGLT showed no significant difference between gender( P> 0. 05),EOFT also showed no significant difference between gender( P>0. 05). Conclusion AGLT is an accurate,resource and time saving method for researching red blood cell storage lesion.
