摘 要:目的探讨成年大鼠脊髓损伤后谷氨酸-天冬氨酸转运体(GLAST)和谷氨酸转运体-1(GLT-1)在胶质细胞表达的变化及其意义。方法健康成年雌性SD大鼠25只,按随机数字表法分为对照组(5只)和实验组(20只),实验组按改良的Allen打击法制作脊髓挫伤动物模型后分为伤后1,3,7及14d组,每组5只。分别于伤后1,3,7及14d应用改良的Tarlov评分评价大鼠后肢神经功能后采用心脏灌注法处死取材,应用免疫荧光染色及计算机图像分析系统(IPP6.0)进行半定量分析,观察GLAST和GLT-1的表达情况。结果(1)单标:对照组GLAST呈中等量表达;实验组伤后1d,GLAST表达略有升高,伤后3,7d呈进行性下降,伤后14d略有升高。伤后3,7和14d,实验组GLAST的表达明显低于对照组(P<0.05)。对照组GLT-1呈中等量表达;实验组伤后1d,GLT-1的表达略有升高,伤后3d表达最少,伤后7,14d略有升高。伤后3,7和14d,实验组GLT-1的表达明显低于对照组(P<0.05)。(2)双标:对照组星形胶质细胞上可见明显GLAST表达;实验组伤后1d,GLAST表达下降,伤后3,7d呈进行性下降,伤后14d开始恢复。伤后3,7d,实验组GLAST与胶质纤维酸性蛋白(GFAP)共表达明显低于对照组(P<0.05)。对照组小胶质细胞上可见少量GLAST表达;实验组伤后1d表达明显上升,伤后3~14d呈进行性上升。伤后3,7和14d,实验组GLAST与OX-42共表达明显高于对照组(P<0.05)。(3)双标:对照组星形胶质细胞上可见明显GLT-1表达;实验组伤后1d表达下降,伤后3,7d呈进行性下降,伤后14d开始恢复。伤后3,7d,实验组GLT-1与GFAP共表达明显低于对照组(P<0.05)。对照组小胶质细胞上可见少量GLT-1表达;实验组伤后1d表达明显上升,伤后3~14d呈进行性上升。伤后1,3,7和14d,实验组GLT-1与OX-42共表达明显高于对照组(P<0.05)。结论大鼠脊髓损伤后GLAST和GLT-1在星形胶质细胞和小胶质细胞的表达模式不同,可能与不同胶质细胞在脊髓损伤及其修复过程中�Objective To investigate the changes and their significance of glutamate-aspartate transporter (GLAST)and glutamate transporter-1(GLT-1)expressions in glial cells following spinal cord injury (SCI)in adult rats.Methods Twenty-five healthy female adult SD rats were randomly divided into control group (5 rats)and experimental group (20rats).The contusive spinal cord injury models were prepared at T10 segment in the rats in the experimental group according to the modified Allen's method.At days 1,3,7 and 14 following SCI,five rats were sacrificed by cardiac perfusion and the spinal cord segments adjacent to the epicenter of injury were obtained at each time point after the neurological function of hind limbs was assessed using the modified Tarlov scale.Changes of GLAST and GLT-1expressions were detected semi-quantitatively using immunofluorescence and computer image analysis system (IPP 6.0).Results (1)Single immunofluorescenee:Moderate GLAST expression was found in the control group.The GLAST expression was increased slightly at day 1after SCI, decreased progressively at days 3and 7after SCI,and increased slightly at day 14after SCI.The GLAST expression in experimental group was significantly lower than those in control group at days 3,7and 14 after SCI (P <0.05).Moderate GLT-1expression was detected in the control group.The expression of GLT-1was increased slightly at day 1after SCI,decreased to the lowest at day 3after SCI,and increased slightly at days 7 and 14 after SCI.The GLT-1expression in experimental group was significantly lower than those in control group at days 3,7 and 14 after SCI (P <0.05).(2)Double immnnofluorescence: GLAST expression was found on astroeytes in the control group.The GLAST expression in experimental group was decreased at day 1after SCI,further decreased progressively at days 3 and 7 after SCI,and started to recover at day 14after SCI.The coexpressions of GLAST and glial fibrillary acidic protein (GFAP)in experimental group were significantly lower than those in the control group at
