摘 要:目的建立凝胶微柱法测定仙茅苷脂质体包封率的方法,并考察影响测定仙茅苷脂质体包封率的因素。方法采用葡聚糖G-50凝胶微柱分离仙茅苷脂质体与未包封仙茅苷,运用紫外分光光度法考察不同内径和柱高的凝胶微柱、不同体积的洗脱液和加样量对包封及未包封药物洗脱行为的影响;采用HPLC法测定仙茅苷含量,计算包封率。结果葡聚糖G-50凝胶微柱的最佳径高比为1∶3.3,洗脱液的体积为0.4 mL,加样量为0.2 mL,此时可将脂质体与游离药物分离开来,平均包封率为69.83%。结论凝胶微柱法适用于仙茅苷脂质体包封率的测定。Objective To establish a method to determine entrapment efficiency of curculigoside liposomes. Methods Sephadex G-50mini-column was used to separate the encapsulated and non-encapsulated curculigoside.The elution behavior of the encapsulated and non-encapsulated drugs were determine by UV spectrophotometry as to the diameter and height of the gel column,the eluent volume and the sample volume. The entrapment efficiency was calculated.Results The optimum diameter-to-height ratio of Sephadex G-50 mini-column was 1:3.3,the eluate volume was 0.4mL,and the sample volume was 0.2mL.At the same time, the free drug was separated from the liposomes and the average entrapment efficiency was 69.83%.Conclusion Sephadex G-50 Mini-column method is simple,rapid,accurate and it is reliable for the determination of the entrapment efficiency of curculigoside liposomes.
