摘 要:目的:观察植物性雌激素柴胡皂苷d(SSd)对大鼠肝星状细胞(HSC-T6)收缩性的影响,并探讨其作用机制。方法:培养大鼠肝星状细胞,分为对照组、内皮素(ET)组、ET+SSd组、ET+SSd+MPP组和ET+SSd+PHTPP组。用胶原收缩法检测雌激素α受体拮抗剂(MPP)和雌激素β受体拮抗剂(PHTPP)对细胞收缩的影响,用Western blot方法在蛋白水平检测雌激素受体表达及Rho激酶通路蛋白的变化。结果:SSd处理后,可明显抑制ET-1引起的胶原凝胶收缩(P<0.05),PHTPP可以部分抵消SSd的抑制作用(P<0.05),MPP作用不明显。Western blot结果显示,各组HSC-T6细胞均表达ERβ,而不表达ERα; ET组Rho A蛋白表达及p-moesin/moesin比值较对照组明显升高(P<0.05),SSd+ET组Rho A蛋白表达及p-moesin/moesin比值较ET组明显降低(P<0.05),SSd+ET+PHTPP组Rho A蛋白表达及p-moesin/moesin比值较SSd+ET组明显升高(P<0.05)。结论:SSd可显著抑制大鼠肝星状细胞收缩; SSd通过ERβ受体抑制大鼠肝星状细胞的收缩,其机制可能与Rho激酶通路的moesin蛋白磷酸化有关。Objective: To observe the effects of plant estrogen Saikosaponin d ( SSd) on the contractility of rat hepatic stellate cells( HSC-T6) ,and investigate its mechanism. Methods: The rat hepatic stellate cells were cultured and divided into the control group,endothelin ( ET) group,ET+SSd group,ET+SSd+MPP group and ET+SSd+PHTPP group. The effects of estrogen α receptor antagonist( MPP) and estrogen β receptor antagonist ( PHTPP) on the cell contraction were detected by collagen contraction method. The proteinexpressions of estrogen receptor and the Rho kinase pathway protein changes were detected by Western blot. Results: After treatment withSSd,the collagen gel contraction induced by ET-1 was significantly inhibited ( P<0.05) . PHTPP could partly counteract the inhibitoryeffect of SSd ( P<0.05) ,and MPP showed no obvious effect. The results of Western blot showed that ER β was expressed in HSC-T6 cellsof all groups,but not ER α; the expression of RhoA protein and the ratio of p-moesin to moesin in the ET group were significantly higherthan those in the control group( P<0.05) ,the expression of RhoA protein and the ratio of p-moesin to moesin in the SSd+ET group weresignificantly lower than those in the ET group( P<0.05) ,and the expression of RhoA protein and the ratio of p-moesin to moesin in theSSd+ ET + PHTPP group were significantly higher than those in the SSd + ET group ( P<0.05) . Conclusion: Saikosaponin d cansignificantly inhibit the contraction of rat hepatic stellate cells via ERβ receptor,and its mechanism may be related to moesinphosphorylation of Rho kinase pathway.
