摘 要:以貉和银狐裘皮制品的毛为研究对象,比较3种DNA提取方法以及针毛和绒毛的不同使用量,并应用荧光定量PCR进行检验。结果表明:3种方法皆可获得较好提取效果,Magen磁珠法略优于AXYGEN法和CTAB-AXYGEN结合法,综合考虑试验成本和提取时间,AXYGEN法的试验成本最低,花费时间最少。在样品的使用量方面,针毛使用15根以上(长度约为5~7 cm),绒毛使用5 mg左右,可达到较好的提取效果。In this study,the hair of raccoon and silver fox fur products were used as samples to compared three kinds of DNA extraction methods,as well as the different number of the guard hair and fine hair,and the fluorescence quantitative PCR was used for testing.The results showed that all the three methods could obtain better extraction effect.Magen beads method was superior to AXYGEN and CTAB-AXYGEN method.However,considering both experimental cost and extraction time,the AXYGEN method had the lowest experimental cost and the least time.In the quantity of the sample used,the guard hair used more than 15 (about 5-7 cm in length ),or the fine hair was about 5 mg,a better extraction effect could be achieved.This study could provide a reliable basis for the selection of DNA extraction program for fur products.
