摘  要:目的探讨硫氢化钠(NaHS)对慢性心力衰竭(CHF)大鼠心肌纤维化及结缔组织生长因子(CTGF)表达的影响。方法 2016年1月—2016年12月于武汉市第一医院无特定病原体(SPF)级动物实验室完成实验。通过腹主动脉缩窄术构建CHF大鼠模型,将SD大鼠随机分为假手术组、模型组、NaHS低剂量组和NaHS高剂量组,每组6只。治疗4周后,采用超声心动图检测每组大鼠心功能,采用HE及Masson染色观察每组大鼠心肌病理形态变化,采用荧光定量PCR(q PCR)和Western blot分别检测各组大鼠心肌组织转化生长因子-β_1(TGF-β_1)和CTGF mRNA及蛋白的表达。结果各组间比较左心室舒张末内径(LVEDD)、左心室收缩末内径(LVESD)、左心室质量/体质量(LVW/BW)、心脏质量/体质量(HW/BW)及左心室射血分数(LVEF)有显著差异(F=15. 898、10. 010、46. 437、21. 520、9. 743,P=0. 004、0. 013、0. 000、0. 000、0. 014)。与模型组比较,NaHS低剂量组和NaHS高剂量组的LVEDD、LVESD、LVW/BW及HW/BW均明显降低(P <0. 05),而LVEF明显升高(P <0. 05),心肌组织病理形态学明显改善,心肌组织中TGF-β_1和CTGF mRNA及蛋白的表达显著降低,且高剂量组的效果优于低剂量组(P <0. 05)。结论NaHS能够降低CHF大鼠的心肌纤维化程度,改善心功能,其作用机制可能与抑制心肌组织中TGF-β_1和CTGF的过度表达有关。Objective To investigate the effects of sodium hydrosulfide (NaHS) on myocardial fibrosis and expression of connective tissue growth factor (CTGF) in rats with chronic heart failure (CHF). Methods The experiment was completed in the Specific pathogen free (SPF) level animal laboratory of Wuhan First Hospital from January 2016 to December2016. The rat models of CHF were established by abdominal aortic constriction. SD rats were randomly divided into sham operation group,model group,low dose of NaHS group and high dose of NaHS group (n = 6 in each group). After 4 weeks of treatment,the cardiac function of each group was detected by echocardiography. The pathological changes of myocardium in each group were observed by HE staining and Masson staining. The mRNA and protein expressions of transforming growth factor-β1 (TGF-β1) and CTGF in myocardial tissues were detected by q PCR and Western blot respectively. Results The comparison of the left ventricular end diastolic diameter (LVEDD),left ventricular end systolic diameter (LVESD),left ventricular weight/body weight (LVW/BW),heart weight/body weight (HW/BW) and left ventricular ejection fraction (LVEF) had significant difference among the groups (F = 15. 898,P = 0. 004,F = 10. 010,P = 0. 013,F = 46. 437,P = 0. 000,F =21. 520,P = 0. 000,F = 9. 743,P = 0. 014). Compared with the model group,the LVEDD,LVESD,LVW/BW and HW/BW in the low dose of NaHS group and high dose of NaHS group were significantly decreased (P < 0. 05),while LVEF was significantly increased (P < 0. 05). The pathological morphology of myocardium was significantly improved in the low dose of NaHS group and high dose of NaHS group. The mRNA and protein expressions of TGF-β1 and CTGF in myocardium were significantly decreased,and the effect of high dose of NaHS group was better than that of low dose of NaHS group. Conclusion The NaHS can reduce myocardial fibrosis and improve cardiac function in CHF rats,and its mechanism may be related to the inhibition of overexpression of TGF-β1 and CTGF in myocar
