摘  要:以实验室保存的鸡致病性大肠杆菌裂解性噬菌体EcP5为研究对象,采用双层琼脂平板法测定最佳感染复数(MOI)、一步生长曲线、热稳定性等生物学特性,研究不同接菌量对扩大培养的影响,分析不同灌服剂量对肠道菌群的影响。结果显示:EcP5的最佳感染复数为0.001;对数生长期在110~150 min之间;在pH7.0~10.0范围内,噬菌体效价稳定;热稳定性较差,40℃作用2 h,滴度下降37.41%,60℃作用20 min噬菌体全部失活;EcP5裂解谱相对较宽,裂解率为40.0%(18/45);扩大培养试验表明,当装液量为70%,接菌量为1.0 mL/100 mL,感染复数为0.001,37℃180 r/min培养3 h,4℃作用24 h时噬菌体效价最高;灌服剂量为0.25 mL/只时,EcP5对鸡肠道菌群和大肠菌群影响最小。本研究为后期的噬菌体治疗提供了理论依据。The laboratory-preserved chicken pathogenic Escherichia coli lytic phage EcP5 was used as the research object. The double-layer agar plate method was used to determine the optimal multiplicity of infection(MOI), one-step growth curve, thermal stability, pH stability and host range included. At the same time, the effect of different inoculation amount on expanded culture, the effects of different dosages on intestinal flora were analyzed. The results showed that the MOI was 0.001, the logarithmic phase was between 110 to 150 min, the titer of EcP5 was stable in the range of pH 7.0 to pH 10.0. Thermal stability of EcP5 was poor, the titer was decreasing by 37.41% at 40 ℃ for 2 h. It would be inactivated completely at 60 ℃ for 20 min. EcP5 had a relatively wide host range, the lysis rate was 40.0%(18/45).The expanded culture test showed that the bacteriophage titer was the highest when the volume of medium was 70%,the inoculum amount was 1.0 mL/100 mL, and the MOI was 0.001, cultured at 37 ℃ 180 r/min for 3 h and kept at 4 ℃ for 24 h. At a dose of 0.25 mL/per chichen, EcP5 had the least effect on the intestinal flora and coliforms. It provided theoretical guidance for the application of phage in the future.
