脑泰方干预的MCAO模型大鼠血浆差异蛋白筛选及其生物信息学分析

摘　　要：目的:应用蛋白质组学技术探索脑泰方干预脑梗死大鼠血浆中的特异蛋白,筛选出药物作用的关键蛋白,为明确中药治疗脑梗死的作用靶点和作用机制研究提供依据。方法:建立MCAO大鼠模型,将脑梗死+脑泰方组、脑梗死组大鼠的血浆采用2-DE技术结合质谱分析进行蛋白质的比较分析,筛选出差异蛋白,运用生物信息学技术对差异蛋白质进行功能分析。结果:(1)对"脑梗死组"血浆与"脑梗死+脑泰方组"血浆的胶图扫描结果进行比较显示:有21个显著差异蛋白点;其中"脑梗死组"血浆中的5个蛋白质(肽)在"脑梗死+脑泰方组"血浆中不存在;另外有13个蛋白质(肽)在"脑梗死+脑泰方组"血浆中比在"脑梗死组"血浆中明显减少,有3个蛋白质(肽)在"脑梗死+脑泰方组"血浆中比在"脑梗死组"血浆中明显增加。结论:通过分析发现脑泰方干预脑梗死的相关特异性蛋白,发现脑泰方可能是通过改善脑血液流变学、血管内皮细胞损伤等途径起作用,阐明中药对于脑梗死可能的治疗靶点和作用机制。Objective:To explore the specific protein in Naotai Formula intervention in rats with cerebral infarction plasma by application of proteomics technology,search the key protein and provide the basis for the research target and mechanism of traditional Chinese medicine in the treatment of cerebral infarction.Methods:The rat model of MCAO was set up.Mass spectrometry analysis was used for plasma protein in cerebral infarction group and cerebral infarction + Naotai Formula group.Using 2-DE technology,the differential proteins were analyzed.Results:(1)Gel map scanning results of cerebral infarction and "cerebral infarction + Naotai Formula" group showed that there were 21 significant difference proteins.The 5 proteins peptide in the plasma of cerebral infarction group(peptide),but those in the "cerebral infarction + Naotai Formula" group did not exist.The other 13 proteins(peptides) in the "cerebral infarction + Naotai Formula" group were significantly lower than that in the cerebral infarction group.There proteins(peptides) in the "cerebral infarction + Naotai Formula" group were significantly higher than that in cerebral infarction group.Conclusion:Through the analysis of proteins related to specific Naotai Formula intervention of cerebral infarction,it found Naotai Formula can cure CI through the role of improving cerebral blood,vascular endothelial cell injury and other ways.And it could be clarified Chinese medicine for the treatment of targets and mechanisms of cerebral infarction.
