摘  要:目的:探讨电针内关预处理对心肌缺血再灌注大鼠炎性因子TNF-α、IL-1β与MKK3/6-p38MAPK通路的影响。方法:将90只大鼠随机分为9组:假手术组(S组)、模型组(M组)、电针预处理组(EA组)、抑制剂预处理组(SB组)、电针+抑制剂预处理组(EA+SB组)、缺血预处理组(IP组)、缺血预处理+抑制剂预处理组(SB+IP组)、药物预处理组(DP组)和药物预处理+抑制剂预处理组(DP+SB组)。进行相应组电针内关穴,治疗结束后,测定大鼠心肌TNF-α、IL-1β的含量,检测各组MKK3/6、p38MAPK、p-p38MAPK的含量。结果:与M组比较,S组、EA组、SB组、EA+SB组、IP、IP+SB、DP及DP+SB大鼠心肌TNF-α、IL-1β及MKK3/6、p38MAPK和p-p38MAPK蛋白的表达明显降低;EA组的上述指标与EA+SB组和SB组比较,显著降低,差异有统计学意义(P<0.05)。EA组的上述指标与IP组和DP组比较,差异没有统计学意义(P<0.05)。结论:p38MAPK信号通路介导了电针内关预处理对I/R大鼠心肌组织的保护作用,并且上游激活物包括参与炎症反应和氧化应激反应的各类相关细胞因子,参与I/R的保护效应。Objective:To investigate the effect of electroacupuncture Neiguan(PC6) at preconditioning on TNF-α,IL-1β and MKK3/6-p38 MAPK pathway in myocardial ischemia-reperfusion rats.Methods:Ninety rats were randomly divided into nine groups:sham operation group,model group,electroacupuncture pretreatment group,inhibitor pretreatment group,electroacupuncture+inhibitor pretreatment group,ischemic preconditioning group,treatment+inhibitor inhibitor group,drug pretreatment group and drug pretreatment+inhibitor pretreatment group.The contents of MKK3/6,p38 MAPK and p-p38 MAPK in the myocardium were measured and the levels of TNF-α and IL-1β in the myocardium were measured.Results:Compared with M group,TNF-α,IL-1β and MKK3/6,p38 MAPK and p were obviously decreased in S group,EA group,SB group,EA + SB group,IP group,IP + SB group,DP group and DP+SB group(P<0.05).The expression of pG38 protein was significantly lower in EA group than that in EA + SB group or SB group(P<0.05),and the difference was statistically significant(P<0.05).There was no significant difference between the EA group and the IP group the DP group(P<0.05).Conclusion:The p38 MAPK signaling pathway mediates the protective effect of electroacupuncture at Neiguan(PC6) pretreatment on myocardial tissue in I/R rats,and upstream activators include various related cytokines involved in inflammatory response and oxidative stress response,and participate in I/R protective effect.
