摘  要:目的探讨心痛宁加味方治疗表柔比星诱导小鼠急性心肌损害的保护作用。方法将60只实验小鼠随机分为6组,10只/组。空白对照组小鼠经腹腔注射生理盐水,模型组小鼠经腹腔内注射表柔比星与生理盐水,丹参组小鼠经腹腔内注射表柔比星与丹参,其他组分别为表柔比星联合低、中、高剂量的心痛宁加味方,处死小鼠后检测脑钠肽(BNP)、心型脂肪酸结合蛋白(h-FABP)、一氧化氮(NO)、超氧化物歧化酶(SOD)活性、肌钙蛋白I(cTnI)水平,同时观察各组小鼠心肌组织的病理形态学改变以及心肌细胞凋亡情况。结果各组BNP、h-FABP、NO、SOD活性、cTnI指标比较,存在显著差异(P <0.05);模型组心肌细胞病例评分明显高于丹参组、低、中、高剂量心痛宁加味方组,并且各组心肌细胞凋亡指数比较存在显著差异(P <0.05);以模型组心肌损害、凋亡效果最为严重,除对照组外以丹参组与高剂量组心肌损害、凋亡最轻。结论心痛宁加味方及丹参对表柔比星诱导的小鼠心肌损害具有良好的保护作用,其可有效降低氧自由基的产生,并通过降低脂质过氧化物含量达到改善心肌损害及调节凋亡的目的。Objective To investigate the protective effect of Xintongning Jiawei Formula on acute myocardial damage induced by epirubicin in mice. Methods Sixty experimental mice were randomly divided into 6 groups with 10 mice in each group. The mice in the blank control group received intraperitoneal injection of saline, the mice in the model group received intraperitoneal injection of epirubicin and saline, and the mice in the Danshen (Radix Salviae Miltiorrhizae) group received intraperitoneal injection of epirubicin and Danshen. Other groups were treated with epirubicin combined with low, medium and high dose Xintongning Jiawei Formula. The brain natriuretic peptide (BNP), cardiac fatty acid-binding protein (h-FABP), nitric oxide (NO), superoxide dismutase (SOD) activity and troponin I (cTnI) levels were detected after the mice were killed. At the same time, the histopathological changes and myocardial cell apoptosis were observed in all groups. Results There were significant differences in the levels of BNP, h-FABP, NO, SOD activity and cTnI among the groups ( P < 0.05); the case score of myocardial cells in the model group was significantly higher than that in the Danshen group, low, medium, and high dose Xintongning Jiawei Formula groups. There were significant differences in apoptotic index of myocardial cells among groups ( P < 0.05). The effect of myocardial damage and apoptosis was the most serious in the model group. Except for the control group, the effect of myocardial damage and apoptosis was the least in the Danshen group and the high dose Xintongning Jiawei Formula group. Conclusion Xintongning Jiawei Formula combined with Danshen has a good protective effect on myocardial damage induced by epirubicin in mice. It can effectively reduce the production of oxygen free radicals, and improve myocardial damage and regulate apoptosis by reducing lipid peroxide content.
