摘 要:目的:研究参附预处理是否对失血性休克大鼠的肺组织起保护作用。方法:雄性SD大鼠随机分为4组(n=10):对照组,失血性休克(HS)组,2mL/kg参附(SF1)组,4mL/kg参附(SF2)组。盐水或参附在在休克前30min通过尾静脉注入大鼠体内。除对照组外,其它3组经10min股动脉缓慢放血使大鼠平均动脉压达(40±5)mmHg诱发失血性休克,通过放血或输血维持大鼠休克状态60min。在复苏后4h,测定大鼠肺组织丙二醛(MDA)、超氧化物歧化酶(SOD)、Bcl-2、Bax和Caspase-3的含量。并测量支气管肺泡灌洗液(BAL)中的蛋白质和细胞含量以及观察肺组织病理学变化。结果:与HS组比较,SF2组MDA、Bax及Caspase-3水平降低;而SOD活性和Bcl-2增高。与此相应的是SF2组BAL中蛋白质和细胞的含量显著降低及肺组织病理学评分降低。结论:大剂量参附预处理对缺血再灌注大鼠肺损伤具有保护作用。涉及的潜在机制是改善肺组织的抗氧化作用。Objective: To study whether the pretreatment of Shenfu, a traditional Chinese medicine, can protect the lung tissue in hemorrhagic shock rats. Methods: Male SD rats were randomly divided into four groups(n=10 per group): control group, hemorrhagic shock group(HS), HS+2 mL/kg Shenfu(SF1) group, and HS+4 mL/kg Shenfu group(SF2). Saline or Shenfu were administered to the rats,then 30 minutes later, hemorrhagic shock was initiated in the experimental groups and this state was maintained for 60 min. Four hours after resuscitation, the malondialdehyde(MDA), superoxide dismutase(SOD), Bcl-2, Bax and Caspase-3 were detected in the lung tissue. The protein and cell contents in bronchoalveolar lavage fluid(BAL) and the pathological changes were also measured. Results:Compared with those in HS group, the levels of MDA, Bax and Caspase-3 decreased in SF2 group,while the activity of SOD and Bcl-2 increased. Correspondingly, the protein and cell contents in BAL and the pathological score were significantly decreased in the lung tissue of SF2. Conclusion: Pretreatment with high-dose Shenfu has a protective effect on lung injury in hemorrhagic shock rats. The potential mechanism involved is improvement of antioxidation.
