摘 要:目的观察经后增殖方对超排卵大鼠卵母细胞分泌因子的影响。方法 30只雌性SD大鼠随机分为空白对照组、模型对照组、中药治疗组。模型对照组和中药治疗组建立超排卵模型,中药治疗组给予经后增殖方颗粒剂灌胃,模型对照组和空白对照组给予生理盐水灌胃,经颈椎脱臼处死后取卵巢组织。采用实时荧光定量PCR(qPCR)和蛋白印迹法(Western Blot)分别检测大鼠卵巢组织中GDF9、BMP15、FGF10mRNA及蛋白的表达。结果模型对照组GDF9、BMP15、FGF10 mRNA及蛋白表达均下降,与空白对照组比较差异具有显著性(P<0.05)。中药治疗组各因子mRNA及蛋白表达水平均高于模型对照组,且差异具有统计学意义(P<0.05)。中药治疗组GDF9、BMP15表达与空白对照组相比差异无统计学意义(P>0.05),FGF10表达显著低于空白对照组(P<0.05)。结论中药经后增殖方能促进超排卵大鼠卵母细胞分泌因子GDF9、BMP15、FGF10 mRNA及其蛋白的表达,推测中药通过提高其表达而改善卵母细胞质量。Objective To observe the effects of Jinghou Zengzhi Recipe(JHZZR) on oocyte secreted factors(OSFs) of rats with controlled ovarian hyperstimulation(COH). Methods 30 SD female rats were randomly divided into blank control group,model control group and traditional Chinese medicine(TCM)group(JHZZR). The models of COH were established in model control group and TCM group. TCM group was given JHZZR granules by gavage, model control group and blank control group were given equal volume saline, then ovarian tissue were collected. The mRNA and protein expression of GDF9, BMP15 and FGF10 in rat ovarian tissues were detected by qPCR and Western Blot,respectively. Results The mRNA and protein expression of GDF9, BMP15 and FGF10 in model control group were decreased, and the difference between blank control group and model control group was significant(P<0.05). Compared with model control group, the mRNA and protein level of GDF9, BMP15 and FGF10 in TCM group were increased, and the difference was statistically significant(P<0.05). There was no significant difference in GDF9 and BMP15 expression between TCM group and blank control group(P>0.05), while the expression of FGF10 was significantly lower than blank control group(P<0.05). Conclusion JHZZR can promote the mRNA and protein expression of GDF9, BMP15 and FGF10 in COH rats, guessing that JHZZR improves oocyte quality by increasing the OSFs’expression.
