摘 要:目的:进一步验证雌激素在骨髓间充质干细胞(BMSC)分化为子宫内膜异位症(EMT)病灶过程中的作用。方法:原代培养BMSC后进行流式细胞分析鉴定,建立小鼠EMT模型并将内异病灶进行HE染色鉴定。将小鼠BMSC经尾静脉注射EMT模型小鼠内,按BMSC是否预处理分为无17β-雌二醇预处理组(对照组)和17β-雌二醇预处理组(预处理组)。1月后EMT病灶切片免疫荧光检测子宫内膜间质细胞(ESC)的B淋巴细胞瘤-2(BCL-2)、增殖细胞核抗原(PCNA)、基质金属蛋白酶-1(MMP-1)蛋白表达差异。结果:小鼠模型EMT病灶HE染色提示建立模型成功。免疫荧光结果发现:预处理组的异位病灶BCL-2的平均积分光密度值高于对照组(56.58±7.07 vs 3.34±0.29),预处理组的异位病灶PCNA的平均积分光密度值高于对照组(83.64±13.07 vs 5.23±0.51),预处理组的异位病灶MMP-1蛋白的平均积分光密度值高于对照组(40.26±7.79 vs 7.64±3.62),差异均有统计学意义(P<0.05)。结论:本实验初步证实17β-雌二醇可能通过促进BMSC迁移、分化,最终促进异位病灶ESC的分化、增殖并减少凋亡,从而增加异位病灶的病变程度。Objective:To evaluate the effect of estrogen on the process of differentiation from bone marrow mesenchymal stem cell(BMSC)into endometriosis(EMT).Methods.Primary BMSC was cultured and flow cytometer was used to identify the cells.EMT mouse model was established followed by HE staining to identify the EMT.BMSC was injected through the tail vein into the EMT mice.The mice was divided into control group and 17 beta estradiol pretreated group according to with or without 17 beta estradiol pretreatment.The immunofluorescence was used to detect the expression of protein BCL-2,PCNA,MMP-1 in endometrial stromal cell(ESC)after one month.Results:The mouse EMT model was proved to be successfully established by HE staining.The immunofluorescence results showed that the mean IOD of BCL-2 in 17 beta estradiol pretreated group was higher than that of control group(56.58±7.07 vs 3.34±0.29).The mean IOD of PCNA in 17 beta estradiol pretreated group was higher than that of control group(83.64±13.07 vs 5.23±0.51 ).The mean IOD of MMP-1 in 17 beta estradiol pretreated group was higher than that of control group(40.26±7.79 vs 7.64±3.62).All the differences were significant(P<0.05).Conclusions..This study had preliminarily confirmed that 17 beta estradiol could increase the degree of EMT through promotion of the migration and differentiation of BMSC and differentiation,proliferation and decreased apoptosis of ESC.
