摘  要:为探究葛根素(Pur)在镉(Cd)致大鼠大脑皮质氧化损伤中的保护作用,40只雄性SD大鼠随机分为4组,分别为对照组、镉组、葛根素组、镉与葛根素共处理组。试验期间,所有大鼠均自由饮用纯净水,每天按200 mg/kg剂量的葛根素对葛根素组、镉与葛根素共处理组大鼠进行灌胃,同时以相同剂量的纯净水将对照组、镉组大鼠进行灌胃,连续处理5周,镉组、镉与葛根素共处理组大鼠在处理4周后连续一周每天用醋酸镉按2.5 mg/kg剂量进行腹腔注射。5周后,分离大鼠大脑皮质,测定大脑皮质内总抗氧化能力(T-AOC),超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性和丙二醛(MDA)的含量。结果表明,与对照组相比,镉组大鼠大脑皮质T-AOC,SOD和GSH-Px活性显著降低(P<0.05),MDA含量显著升高(P<0.05);与镉组相比,镉与葛根素共处理组T-AOC,SOD和GSH-Px活性显著升高(P<0.05),MDA含量显著降低(P<0.05)。说明葛根素在镉致大鼠大脑皮质氧化损伤中具有一定的保护作用。In order to evaluate the oxidative damage induced by cadmium exposure in cerebral cortex of SD rats and protective effects of puerarin,forty male SD rats(70±5) g were randomly divided into four groups:control group,Cd-treated group,puerarin group,Cd and puerarin co-treated group.All rats were free acess to purified water during the experiment.When puerarin(200 mg/kg) was given to rats by gavage in the puerarin group and the co-treated group,the same dose of purified water was given to rats by gavage concurrently in the control group and the Cd-treated group over 5 weeks.The Cd Ac2(2.5 mg/kg) was given to rats by intraperitoneal injection daily over 1 week at the fifth week in the Cd group and the co-treated group.Five weeks later,the cerebral cortex was isolated.The T-AOC,the activities of SOD and GSH-Px and the contents of MDA in cerebral cortex were measured.In result,compared with the control group,the T-AOC,the activities of SOD and GSH-Px in cadmium-exposed groups all decreased significantly(P<0.05),and the contents of MDA in cerebral cortex increased significantly(P<0.05).Compared with the cadmium group,the T-AOC,the activities of SOD and GSH-Px in co-treated groups all increased significantly(P<0.05),and the contents of MDA decreased significantly(P<0.05).The results indicated that puerarin can protect the oxidative damage caused by cadmium in cerebral cortex of rats.
