摘  要:基于超高效液相色谱-四极杆飞行时间高分辨质谱(UHPLC-QTOF-MS/MS)联用技术,对栽培两面针不同部位(根、茎、枝)的生物碱进行比较分析,并对其中的氯化两面针碱,白屈菜红碱进行定量,为栽培两面针的质量控制提供依据。色谱柱:ACQUITY UPLC BEH C18(2.1×100 mm,1.7μm),流速0.5 mL/min,柱温35℃,检测波长为272 nm,流动相:A(0.1%甲酸-水),B(乙腈),梯度洗脱,PDA检测器,ESI正离子模式采集。根据获得的母离子和二级碎片离子精确质量数信息,结合对照品及文献,从两面针根、茎、枝中鉴定了37个成分,其中,30个成分为共有成分。4年生栽培两面针根部、茎、枝氯化两面针碱的含量范围为0.12%~0.23%,白屈菜红碱的含量范围为0.06%~0.15%:该方法分辨率好、灵敏度高,可用于两面针药材的定性定量分析,有助于对该药材的品质进行深入研究。To qualitatively and quantitatively analyze the alkaloids in different part of cultivated Zanthoxylum nitidu by UHPLC-QTOF-MS/MS,the chemical constituents were separated on a column of ACQUITY UPLC BEH C18(2.1× 100 mm,1.7 μm)with a flow rate of 0.5 mL/min at 35 ℃, gradient elute was performed with a mobile phase of A (0.1% formic acid) and B (acetonitrile), PDA detector was set at 272 nm. QTOF-MS was set on the positive mode in order to obtain a stronger response signal. A total of 37 alkaloids and other compounds were unequivocally or tentatively identified from cultivated Z. nitidum by comparing their retention times, UV spectra and HR-MS data with those of the reference substances and the data of the literatures. Among them, 30 compounds were found in the root, stem and branch of Z. nitidum. The content of nitidine chloride in different part of Z. nitidum ranged from 0.12% to 0.23% and the content of chelerythrine ranged from 0.06% to 0.15%. It is proposed that the methods described here can be applied for rapid evaluation, quality control and authenticity establishment of Z. nitidum.
