摘 要:目的探讨柴胡皂苷D对乳腺癌MCF-7细胞株内质网应激(ERS)所致的凋亡及Calnexin/Calreticulin循环的影响。方法将乳腺癌MCF-7细胞分为对照组、低、中、高剂量组;检测各组细胞增殖、迁移及凋亡情况;Western blot检测各组细胞Calnexin、Calreticulin、GRP78、CHOP的表达。结果与对照组相比,低、中、高剂量组细胞增殖率、迁移能力降低,细胞凋亡率增加,且细胞中Calnexin、Calreticulin、GRP78、CHOP蛋白表达增高。结论柴胡皂苷D可能通过影响MCF-7细胞Calnexin/Calreticulin循环,加重乳腺癌细胞ERS,引发细胞凋亡,从而降低细胞增殖率及迁移能力。Objective To investigate the effect of Saikosaponin D on Calnexin/Calreticulin circulation and endoplasmic reticulum stress (ERS)induced apoptosis in MCF-7 cell line of breast cancer.Methods MCF-7 cells are divided into control group,low,middle and high dose group.The cell proliferation rates, cell scratch assay and cell apoptosis of all groups is detected.Western blot is used to detect the expression of Calnexin,Calreticulin,GRP78 and CHOP.Results Compared with the control group,the cell proliferation rate and migration ability of the low,middle and high dose group decrease,and the apoptosis rate increase.And the expression of Calnexin,Calreticulin,GRP78 and CHOP protein increased in low,middle and high dose groups. Conclusion Saikosaponin D may reduce cell proliferation rate and migration by affecting the Calnexin / Calreticulin cycle of MCF-7 cells,increasing ERS in breast cancer cells and inducing cell apoptosis.
