摘  要:目的研究白细胞介素-1β(IL-1β)在心房神经节丛(GP)注射后对其功能及神经活性的影响。方法 12只健康成年雄性家犬随机分为2组。实验组(n=6)在右前GP(ARGP)注射0.1ml IL-1β溶液(20μg/ml);对照组(n=6)注射等量生理盐水。分别在基础状态(注射前)和注射10min后,以高频电刺激诱导窦性心率(sinus rate,SR)降低,测定ARGP功能;使用微电极记录神经放电,测定ARGP神经放电的频率和振幅。结果实验组注射10min后,电刺激ARGP诱导的SR减慢作用与基础状态相比明显增强[以刺激电压梯度3(3~5V)为例,(48.7±11.2)%vs(35.1±7.8)%,P<0.05],对照组未见明显改变(P>0.05);实验组注射10min后ARGP神经活性频率和振幅均显著上升[频率:(69±9)次/分vs(32±5)次/分;振幅:(0.25±0.06)mV vs(0.12±0.05)mV,P<0.05],对照组注射前后无显著差异(P>0.05)。结论 IL-1β对ARGP的功能及神经活性具有显著的激活作用。Objective To investigate the effects of Interleukin-1β (IL-1β) microinjection on the function and neural activity of atrial ganglionated plexi (GP). Methods Twelve healthy adult male dogs were randomly divided into two groups.IL-1β (20μg/ml,0.1 ml) (the experimental group,n=6) or saline (the control group,n=6) was microinjected into anterior right GP (ARGP).High frequency stimulation (HFS)was delivered and the relative change of sinus rate (SR)were measured as GP function.Microelectrodes were used to record neural spikes,determining the frequency and amplitude of ARGP nerve discharge.The function and neural activity of ARGP were measured at baseline (before microinjection)and 10 minutes after microinjection. Results Compared to baseline,IL-1β microinjection increased the SR slowing effects induced by HFS of ARGP in the experimental group.Taking the stimulation voltage gradient 3 (3~5 V) as an example, (48.7±11.2)% vs (35.1±7.8)%,P<0.05.However,there were no significant difference in the control group.IL-1β microinjection also significantly increased the frequency and amplitude of neural spikes recorded from ARGP in the experimental group[(69±9)beats/min vs (32±5)beats/min; (0.25±0.06)mV vs (0.12±0.05)mV,P<0.05],whereas no distinct changes were shown in the control group. Conclusion IL-1β microinjection observably enhances the function and neural activity of ARGP.
