益气活血组方对大鼠骨髓源性内皮祖细胞增殖、迁移和黏附的影响

摘　　要：目的:探究内皮祖细胞(endothelial progenitor cell,EPCs)的原代培养及鉴定方法,并探讨益气活血组方对大鼠骨髓源性内皮祖细胞增殖、迁移和黏附的影响。方法:将EPCs随机分为空白血清组、5%、10%、15%含药血清组和对照组,以相应血清干预原代EPCs,并分别通过CCK8法、Transwell迁移实验、黏附实验评估对EPCs的增殖、迁移、黏附的影响。结果:空白血清组和含药血清组EPCs增殖、迁移、黏附能力均高于对照组(P<0.05或P<0.01)。10%、15%含药血清组EPCs增殖、迁移、黏附能力均高于空白血清组(P<0.05);10%、15%、5%含药血清组与空白血清组比较,差异均无统计学意义(P>0.05)。结论:益气活血组方能提高大鼠骨髓源性内皮祖细胞增殖、迁移和黏附能力,且与含药血清浓度值呈正相关,并且血清浓度的平台值为15%,超过这个阀值时,血清浓度的增高对细胞的生长影响不大。Objective:To explore the primary culture and identification methods of EPCs,and to observe the effect of Yiqi Huoxue Prescription (益气活血方,YQHXP)on rat bone marrow derived endothelial progenitor cell proliferation,migration and adhesion.Methods:EPCs were randomly divided into blank serum group,5%, 10% and 15% drug-containing serum group and control group.The primary EPCs were intervened with corresponding serum.The effects on EPCs proliferation,migration and adhesion were evaluated by CCK8 method, Transwell migration test and adhesion test,respectively.Results:The proliferation,migration and adhesion of EPCs in blank serum group and drug-containing serum group were higher than those in control group (P<0.05 or P<0.01).EPCs proliferation,migration and adhesion ability in 10% and 15% drug-containing serum group were higher than those in blank serum group (P<0.05);there was no significant difference between 10% drugcontaining serum group and 15% drug-containing serum group and 5% drug-containing serum group and blank serum group (P>0.05).Conclusion:Yiqi Huoxue prescription can improve the proliferation,migration and adhesion ability of rat bone marrow-derived endothelial progenitor cells,which is positively correlated with the concentration of drug-containing serum,and the plateau value of serum concentration is 15%.When the plateau value exceeds this threshold,the increase of serum concentration has little effect on cell growth.
