摘  要:目的:观察脑低灌注损伤后大鼠海马组织NLRP3及其下游分子Caspase-1的表达,探讨补阳还五汤的影响及其作用机制。方法:双侧颈总动脉阻断建立脑低灌注模型,HE染色观察海马病理组织学改变,免疫组化法测定海马NLRP3、Caspase-1的水平。结果:脑低灌注导致大鼠海马神经细胞数量减少,核萎缩,细胞排列松散,间隙变大,排列不规则;补阳还五汤可有效改善脑低灌注所致的海马损伤。模型组NLRP3、Caspase-1的表达明显高于正常组(P<0.01);补阳还五汤组大鼠海马组织NLRP3、Caspase-1的表达明显低于模型组(P<0.05)。结论:补阳还五汤可能通过抑制脑低灌注中NLRP3炎性小体的激活发挥脑保护作用。Objective:To observe the expression of NLRP3and its downstream molecule Caspase-1 in hippocampus of rats after cerebral hypoperfusion injury,and to explore the effect of Buyang Huanwu Decoction (补阳还五汤,BYHWD)and its mechanism.Methods:Brain hypoperfusion model was established by bilateral common carotid artery occlusion.Histopathological changes of hippocampus were observed by HE staining.The levels of NLRP3 and Caspase-1 in hippocampus were measured by immunohistochemistry.Results:Hypoperfusion leads to a decrease in the number of hippocampal neurons,nuclear atrophy,loose cell arrangement,enlarged gap and irregular arrangement in rats.BYHWD can effectively improve hippocampal injury caused by cerebral hypoperfusion.The expression of NLRP3 and Caspase-1 in the model group was significantly higher than that in the normal group (P<0.01);the expression of NLRP3 and Caspase-1 in the hippocampus of rats in BYHWD group was significantly lower than that in the model group (P<0.05).Conclusion:Buyang Huanwu Decoction may play a protective role by inhibiting the activation of NLRP3 inflammatory corpuscles in cerebral hypoperfusion.
