摘 要:目的了解宁波市2014-2017年麻疹病毒野毒株的基因型特征。方法采集宁波市2014-2017年疑似麻疹病人咽试子标本,采用荧光定量RT-PCR检测麻疹病毒核酸,采用Vero/SLAM细胞进行麻疹病毒分离,采用RT-PCR扩增分离株核蛋白基因,对扩增产物进行测序和比对分析。结果共分离获得麻疹病毒株79株,其中71株为H1a基因亚型,8株为B3.1基因亚型。H1a基因亚型分离株之间核苷酸和氨基酸同源性分别为96.76%-100%和96.57%-100%;与H1a亚型参考株China93-2的同源性分别为97.49%-98.88%和95.86%-99.32%;与中国S191疫苗株的同源性分别为89.91%-91.70%和86.26%-89.32%。B3.1基因型分离株之间核苷酸和氨基酸同源性分别为98.63%-100%和98.64%-100%;与B3.2基因亚型参考株的同源性分别为95.61%-96.82%和95.61%-96.82%;与B3.3基因亚型参考株的同源性分别为97.50%-97.97%和95.89%-97.28%;与GenBank中同时期其他地区上传的B3.1基因亚型序列的同源性分别为95.52%-100%和98.65%-100%;与中国S191疫苗株的同源性分别为87.13%-88.64%和93.22%-91.95%。结论 2014-2017年宁波市麻疹病毒的优势基因型为H1a基因亚型,输入基因型可引起小范围流行,应加强麻疹的病原学监测。Objective To determine genetic characteristics of measles viruses isolated in Ningbo city during 2014-2017. Methods We collected throat swab specimens from suspected measles cases during 2014-2017 to detect measles virus nucleic acid by fluorescent quantitative RT-PCR to isolate measles virus by Vero/SLAM cell culture and amplify nucleoprotein gene of isolates by RT-PCR for a sequencing analysis of amplification products. Results Seventy-nine measles virus strains were isolated; 71 were H1 a subgenotype, and 8 were B3.1 subgenotype. Homologies of nucleotides and amino acids within H1 a subgenotype isolates were 96.76%-100% and 96.57%-100%, respectively; between H1 a subgenotype isolates and the China93-2 reference strain were 97.49%-98.88% and 95.86%-99.32%; and between H1 a subgenotype isolates and the Chinese S191 vaccine strain were 89.91%-91.70% and 86.26%-89.32%. Homologies of nucleotides and amino acids within B3.1 genotype isolates were 98.63%-100% and 98.64%-100%, respectively; between B3.1 genotype isolates and the B3.2 subgenotype reference strain were 95.61%-96.82% and 95.61%-96.82%; between B3.1 genotype isolates and the B3.3 subgenotype reference strain were 97.50%-97.97% and 95.89%-97.28%; between B3.1 genotype isolates and other B3.1 genotype strains simultaneously uploaded to GenBank were 95.52%-100% and 98.65%-100%; and between B3.1 genotype isolates and the S191 vaccine strain were 87.13%-88.64% and 93.22%-91.95%. Conclusions The H1 a subgenotype of measles virus was the predominant genotype during 2014-2017 in Ningbo, with small epidemics caused by imported genotypes. We should strengthen pathogenic monitoring of measles.
