摘  要:目的探讨黄芩素(Bai)对小鼠肠缺血再灌注(I/R)致急性肾损伤的作用及机制。方法小鼠分为4组:假手术组、I/R组、Bai+I/R组、Bai+锡原卟啉Ⅸ(Sn PP)+I/R组。Bai+I/R组和(Sn PP)+I/R组于缺血前1 h腹腔给予100 mg/kg的Bai,(Sn PP)+I/R组于缺血前30 min腹腔给予50 mg/kg的Sn PP。1 h后恢复血流,再灌注4 h后处死小鼠。观察小鼠肾脏组织病理学变化,检测血清尿素氮(BUN)、肾组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、肾脏组织细胞凋亡指数,检测肾脏组织血红素氧合酶-1(HO-1) mRNA和蛋白水平。结果 Bai+I/R组肾脏HO-1的mRNA及蛋白水平均高于I/R组,肾损伤程度减轻,血清BUN水平和肾组织MDA含量降低、SOD活性升高、细胞凋亡指数明显降低。Bai+Sn PP+I/R组肾损伤程度增高,血清BUN和肾组织MDA含量升高,细胞凋亡指数明显增加。结论 Bai预处理可以有效地减轻小鼠肠I/R致急性肾损伤。Objective To investigate the effect and mechanism of Baicalein (Bai)on acute renal injury induced by intestinal ischemia-reperfusion(I/R)in mice.Methods Mice were divided into sham operation group,I/R group,Bai +I/R group, Bai +tin protoporphyrin IX (SnPP)+I/R group.100mg/kg Bai was given to Bai +I/R group and (SnPP)+I/R group by abdominal cavity at 1h before isehemia,while (SnPP)+I/R group was given 50mg/kg SnPP by abdominal cavity at 30min before ischemia.After !h,the blood flow was restored and the mice were killed after reperfusion for 4h.To observe the pathological changes of mouse renal tissue,and the serum urea nitrogen(BUN),renal tissue malondialdehyde(MDA)content, superoxide dismutase(SOD)activity and apoptosis index of kidney tissue,heme oxygenase-1(HO-1)mRNA and protein levels in kidney tissue were detected.Results mRNA and protein levels of HO -1in Bai +I/R group were all higher than those in I/R group;the degree of renal injury reduced,serum BUN level and MDA content in renal tissue decreased,SOD activity .increased,and apoptotic index decreased significantly.In Bai +SnPP +I/R group,the degree of renal injury increased, the content of serum BUN and renal tissue MDA increased,and the apoptosis index increased significantly.Conclusion Bai pretreatment can effectively reduce the acute renal injury induced by I/R in mouse intestine.
