基 金:芦荟提取物抗光老化机制研究及其在护肤品方面的应用(校级课题:K2016-32)
摘 要:采用正交试验法优化超临界CO2法萃取中华芦荟的条件及研究萃取物的抗氧化活性。以乙醇夹带剂的浓度、萃取温度和萃取压力为考察因素,总酚的提取率作为考察指标,用L9(33)正交表优化工艺条件,福林酚法测定萃取物总酚含量,用DPPH法评估萃取物的抗氧化活性。通过单因素和正交试验结果分析得到,中华芦荟超临界CO_2萃取最优条件为:乙醇夹带剂的质量分数60%,萃取温度40℃,萃取压力30 MPa,在此条件下相对于没食子酸的萃取物总酚质量分数为4.83%,DPPH的半数清除浓度为(0.19±0.02)mg/mL。该优选的工艺条件明显提高超临界CO_2萃取中华芦荟总酚的提取效率从而增强了其抗氧化活性。The extraction process of Aloe chinensis Baker was optimized by orthogonal test,and the antioxidant activities of obtained extracts were explored.With extraction rate of total polyphenols as the evaluation index,the concentration of ethanol entrainer,extraction pressure and extraction temperature as investigation factors,the process conditions were optimized with L9 (33) orthogonal table,the content of total polyphenols was determined by Folin-Ciocaileu method.The antioxidant activity was assessed by DPPH method.The results showed that through single factor experiments and orthogonal test,the optimal conditions for the extraction of Aloe chinensis Baker by supercritical CO2extraction were obtained as follows:the mass ratio of ethanol entrainer was 60%,extraction temperature was 40℃,and extraction pressure was 30 MPa.Under these conditions,the content of total polyphenols was 4.83% relative to gallic acid,the half scavenging concentration (IC50)towards DPPH was (0.19±0.02) mg/mL.The optimized process conditions of this experiment significantly improved the extraction efficiency of the total polyphenols from Aloe chinensis Baker,also its antioxidant activity was enhanced.
