摘 要:为对海南陵水地区自然发病的幼龄暹罗鳄进行病原菌分离鉴定,本研究采用形态与生理生化特性测定并结合16S r RNA基因系统发育学分析方法,对分离的LS5菌株进行鉴定;同时对分离株进行药物敏感试验及对小鼠致病性试验。结果显示,该菌呈革兰氏阴性杆菌,氧化酶等生理生化特性与简达气单胞菌基本一致;根据16S r RNA基因的系统进化及其同源性分析,可确定分离的菌株LS5与简达气单胞菌属于同一系统发育分支,其基因与FJ940830.1 Aeromonas jandaei p W23菌株同源性高达100%;LS5菌株对小鼠的半数致死量为4.1×10~5 cfu/只,具有较强致病性;该菌对庆大霉素、丁胺卡那霉素、环丙沙星敏感,对阿莫西林、利福平、氨苄西林等抗生素耐药。本研究从海南发病幼鳄体内首次发现简达气单胞菌,同时检测到其具有较强致病性,通过药敏试验为该病的防治奠定基础。A pathogenic bacteria,designated LS5,were isolated fi:om the young Siamese crocodile in Lingshui area of Hainan province and identified by morphological,physiological and biochemical characteristics combined with the 16S rRNA phylogenetic analysis.At the same time,the isolate was tested for drug susceptibility and pathogenicity in mice.The results showed that the bacteria was gram-negative bacilli,oxidase and other physiological and biochemical characteristics were basically consistent with that of Aeromonas.According to the phylogenetic and homology analysis of the 16S rRNA gene,the isolate LS5 and Aeromonas were identified as the same phylogenetic branch,and the homology was 100%to A.jandaei PW23strain.The LDs0of the isolate LS5was 4.1×105cfu,indicating the isolate was a strong pathogenic to mice.The isolate was sensitive to gentamicin,amikacin, ciprofloxacin,and resistance to amoxicillin,rifampicin and ampicillin and other antibiotics.In this study,A.jandaei was first found in the infected young Siamese alligator in Hainan,which laid a foundation for the prevention and treatment of the related disease.
