摘 要:目的观察补中益气颗粒对实验性自身免疫性甲状腺炎(EAT)大鼠血清甲状腺功能、甲状腺自身抗体和组织病理形态的影响。方法运用猪甲状腺球蛋白和弗氏佐剂混合免疫注射法,联合高碘喂养法复制EAT大鼠模型,随机分为模型组、补中益气组、优甲乐组和补中益气+优甲乐组,用药干预2个月后,酶联免疫吸附法(ELISA)测定大鼠外周血游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、血清促甲状腺素(TSH)、抗甲状腺过氧化物酶抗体(TPOAb)、甲状腺球蛋白抗体(TGAb)水平,HE染色法光镜下观察大鼠甲状腺组织病理改变。结果与空白组比较,模型组大鼠FT3、FT4、T3、T4水平显著升高(P <0. 05),TPOAb、TGAb水平显著升高(P <0. 05),甲状腺组织淋巴细胞的浸润明显;与模型组比较,补中益气组大鼠FT3、FT4、T3、T4水平显著降低(P <0. 05),补中益气+优甲乐组大鼠FT3、T3、T4水平显著降低(P<0. 05),用药3组大鼠血清TPOAb、TGAb水平显著降低(P <0. 05)。光镜下用药3组大鼠甲状腺组织淋巴细胞的浸润程度较模型组明显减轻,浸润程度改善最明显的是补中益气+优甲乐组,其次是补中益气组、优甲乐组。结论补中益气颗粒能够改善EAT大鼠甲状腺功能,降低甲状腺抗体滴度,减轻大鼠甲状腺组织病理损害,改善免疫功能。Objective To investigate the effects of Buzhong Yiqi Granule( Center-supplementing and Qiboosting,BZYQG) on thyroid function,thyroid autoantibodies and histopathologic morphology in rats with experimental autoimmune thyroiditis( EAT). Methods Animal models of EAT was established by intravenous injection of thyroglobulin mixed with Freund’s adjuvant immunization,and feeding with high iodine water. All EAT rats were randomly divided into model group, BZYQG group, Euthyrox( Levothyroxin Sodium Tablet or L-T4) group and BZYQG + Euthyrox combination treatment group. After2 months of treatment, FT3, FT4, T3, T4, TSH, TPOAb, and TGAb in peripheral blood and pathological changes in thyroids were observed by using enzyme-linked immunosorbent assay( ELISA)and HE staining respectively. Results The levels of FT3,FT4,T3,T4,TPOAb and TGAb were significantly increased in the model group compared with blank group( P < 0. 05),and lymphocyte infiltration in thyroid tissue was more apparent in the model group compared with blank group. Compared with model group,the levels of FT3,FT4,T3,T4 decreased significantly( P < 0. 05),and the level of TSH had no statistical significance( P > 0. 05) in BZYQG group. Comparison of thyroid function between BZYQG group and combination treatment group presented differences with no statistical significance( P >0. 05). The levels of TPOAb and TGAb were significantly reduced in BZYQG group compared with the model group( P < 0. 05). Comparison of TPOAb and TGAb among BZYQG group,Euthyrox group and combination treatment group presented with no statistical significance( P > 0. 05). Compared with model group,the degree of lymphocyte infiltration in thyroid tissue decreased significantly in three intervention groups,and the mixed medicine group was the most obvious one,followed by BZYQG group and L-T4 group. Conclusion BZYQG has the effects of maintaining thyroid functions,reducing thyroid antibody titer,relieving thyroid histopathological damage and improving autoimmunity.
