摘 要:刚地弓形虫病作为一种人畜共患病严重影响人体健康和畜牧业发展,但目前仍无有效的治疗药物,因而疫苗的研制尤为重要。弓形虫侵入宿主细胞是其繁殖、致病的重要环节。在这个过程中微线体蛋白先形成复合体对宿主细胞进行粘附,然后与棒状体颈部蛋白形成移动连接,在Ca2+以及相关激酶、蛋白的调节下虫体依赖肌动-肌球蛋白马达驱动其侵入宿主细胞,这不仅是虫体增殖所必需,也是虫体免疫逃避及慢性感染的重要手段,因而如何阻止/减缓弓形虫侵入宿主细胞成为疫苗研制的重要切入点。本文将从弓形虫侵入过程涉及到的相关蛋白入手,对其疫苗的研究进展进行综述。Toxoplasmosis is a zoonosis that severely impacts human health and the development of animal husbandry,but there is still no effective treatment for it,so the development of a vaccine is particularly important.Invading host cells are an important part of T.gondii’s reproduction and pathogenicity.T.gondii invades host cells with a unique sliding motion.A microneme protein first forms complexes to adhere to the host cell and it then forms a moving junction with the rhoptry neck protein.Regulated by Ca2+and related kinases and regulatory proteins,T.gondii relies on a myosin motor that propels them to invade host cells.At the same time,the rhoptry protein secreted by T.gondii participates in the formation and expansion of a parasitophorous vacuole(PV),which also promotes the entry of T.gondii into the host cell.Finally,a rhomboid protease(TgROMs)cuts off the junction area and completes the invasion.T.gondii proliferates in the PV and finally bursts cells,resulting in acute dissemination,or it forms cysts in tissues that cause chronic infection,which is conducive to the survival and development of the parasite.Thus,T.gondii invasion of host cells is not only necessary for its proliferation but also an important means for immune evasion and chronic infection.Therefore,preventing or slowing down the invasion of host cells by T.gondii is an important approach for vaccine development.With the development of new antigens,adjuvants,and immunization strategies,potential vaccines against T.gondii range from recombinant antigens(single,binding,or chimeric)to synthetic peptides and nucleic acids.Some researchers have conducted preliminary studies on the protective effect of protein vaccines against T.gondii invasion,and they have obtained some results.This paper will review advances in the study of vaccines against the proteins involved in T.gondii invasion.
