摘  要:为了探讨淋巴细胞白血病缺失基因1 (deleted in lymphocytic leukemia 1, DLEU1)的功能及其在膀胱癌发展进程中相关联的基因及信号通路,采用Atlas of Genetics and Cytogenetics in Oncology and Haematology、Dcode、NCBI、GWAS Catalog、Targetvalidation、FireBrowse、Wanderer、GEPIA、Multi Experiment Matrix和LinkedOmics等综合分析DLEU1基因的功能。结果显示:DLEU1基因定位于人13q14.2~q14.3且高度保守;该基因广泛表达于人体多种正常组织,并与身高、原发性胆汁性肝硬化等生物学表型相关;在膀胱癌等多种肿瘤中广泛高表达,且其高表达与膀胱癌总体生存率和无病生存率的显著降低相关。此外,基因信号通路富集分析结果显示,在膀胱癌等多种肿瘤中DLEU1均与ncRNA代谢途径密切相关。以上结果提示, DLEU1可能是膀胱癌潜在的癌基因和治疗靶点,其主要通过调节ncRNA代谢途径参与膀胱癌的进程。To explore the function of deleted in lymphocytic leukemia 1 (DLEU1) and its related genes and signaling pathways in bladder cancer progression by bioinformatics analysis, many tools, including Atlas of Genetics and Cytogenetics in Oncology and Haematology, Dcode, NCBI, GWAS Catalog, Targetvalidation, FireBrowse, GEPIA, Multi Experiment Matrix and LinkedOmics, were applied to comprehensively analyze the function of the DLEU1 gene. The gene is located on human chromosome 13q14.2~q14.3 and highly conserved in different species. It is widely expressed in many normal human tissues, and is associated with body height, primary biliary cirrhosis and other biological phenotypes. It is also widely expressed in bladder cancer and many other tumors. Its high expression in bladder cancer is associated with a significant decrease in both overall and disease-free survival rates. The enrichment analysis further found that DLEU1 is closely related to ncRNA metabolic pathways in many tumors including bladder cancer. DLEU1 may be a potential oncogene and therapeutic target for bladder cancer. It is mainly involved in the progression of bladder cancer by regulating ncRNA metabolic pathways.
