摘 要:目的对1株海洋真菌Penicillium janthinellum HK1-6进行发酵优化,提高其代谢产物青霉酸的产量,以期获得足量的化合物研究青霉酸对水稻纹枯病菌的拮抗性。方法采用正交实验设计对真菌HK1-6产青霉酸的土豆液体培养基进行优化,采用高效液相色谱法对青霉酸产量进行定量分析。结果研究表明,真菌HK1-6高产青霉酸的土豆液体培养基各因素最优水平为葡萄糖30g·L^(-1),土豆200g·L^(-1),海盐10g·L^(-1),pH为4,产量可达0.908g·L^(-1),而原发酵条件下青霉酸产量约为0.423g·L^(-1)。结论真菌HK1-6采用最优土豆液体培养基发酵后,青霉酸的产量显著提高,产量较原培养条件提高约1.1倍。Objective To increase the yield of penicillic acid produced by the marine-derived fungus Penicillium janthinellum HK1-6 by optimizing the fermentation conditions for further studying about antifungal effects of penicillic acid on rice sheath blight pathogen(Rhizoctonia solani).Methods Orthogonal experimental design was used to optimize the fermentation conditions for the fungus HK1-6 cultured by potato dextrose liquid medium,and high performance liquid chromatography(HPLC)was used to analyze the yield of penicillic acid.Results The optimum fermentation condition of potato dextrose liquid medium was potato 200 g·L-1,dextrose 30 g·L-1,sea salt 10 g·L-1,pH=4,and the production yield of penicillic acid was up to 0.908 g·L-1,while the production yield was 0.423 g·L-1 using the originalculture medium.Conclusion When the fungus HK1-6 were cultured by optimum fermentation condition,the yield of penicillic acid increased obviously and was up to about 1.1 times compared to the initial culture.
