摘 要:结合六倍体小麦品系P7001与P216杂交F5代群体中1对相对性状池的二代转录组测序的单核苷酸多态性(SNP)分析结果,应用最近开发的CAPS/dCAPS Designer工具进行批量的CAPS/dCAPS(衍生/限制性内切酶多态性)标记开发,最终筛选20个CAPS/dCAPS标记进行亲本间的多态性验证。结果表明,将二代转录组测序的SNP分析结果和高通量的CAPS/dCAPS标记设计软件相结合能高效地进行多态性分子标记的开发。The single nucleotide polymorphism(SNPs)from the transcriptom sequencing of two pools with contrasting traits in lesion mimic were analyzed,and these two pools were derived from the population of the cross of P7001 and P216(F5).The CAPS/dCAPS(derived/cleaved amplified polymorphic sequences)markers were designed in a high-throughput way using the software CAPS/dCAPS Designer.Twenty CAPS/dCAPS markers polymorphic between the two pools were validated in the parental wheat lines.These results showed that application of the CAPS/dCAPS marker design tool for SNPs from transcriptom sequencing is efficient in the design of polymorphic molecular markers for wheat with complex genomes.
