摘 要:目的利用公共基因芯片数据库GEO、中药有效成分数据库BATMAN-TCM、生物信息分析平台GCBI等在线资源对上市抗风湿苗药黑骨藤追风活络胶囊已知有效成分进行分析,预测分析该药治疗人类类风湿关节炎(Rheumatoid arthritis,RA)的潜在效应靶基因和通路。方法从BATMAN中药化合物数据库搜寻苗药黑骨藤追风活络胶囊(黑骨藤,追风伞,青风藤)所含有效成分作用的效应基因数据集Ⅰ;从GEO筛选符合纳入条件的人RA滑膜组织全基因组RNA芯片(样本为RA患者和正常人的滑膜组织)利用GCBI在线实验室筛选出RA与正常组织的差异基因数据集Ⅱ;分别对上述两个数据集及两者的交集通过GCBI进行GO富集分析、KEGG通路分析以及参与生物功能的蛋白相互作用网络分析(PPI分析)。结果从符合研究纳入条件的3块RA患者与常人滑膜组织基因芯片数据集最终得到455个差异性基因,从BATMAN中药化合物数据查到黑骨藤追风活络胶囊的药物组分调节基因432个,交集得到共27个潜在效应靶基因,后续生信分析及文献回溯证实其中STAT1、MAPK1、RAC2、CDK1、PRKCB与周围基因蛋白互作关系较为突出,STAT1-ERKs相关通路可能是黑骨藤追风活络胶囊抗RA潜在作用方向。结论利用生物信息学有助于黑骨藤追风活络胶囊成分针对RA滑膜组织的治疗效应靶基因探索,为进一步的实验研究指明方向,提供理论依据。OBJECTIVE Using public databases(including GEO,BATMAN-TCM) to make bioinformatics analysis to explore the target gene of Miao Medicine HeiGuTeng ZhuiFeng HuoLuo capsule acting on rheumatoid arthritis.METHODS Herbal ingredients and targeting genes named Data set Ⅰ were collected from BATMAN-TCM Database,Gene microarray profiles on RA(samples of synovial membranes came from rheumatoid arthritis patients and healthy donors) were got from GEO(Database of Gene Expression Omnibus),then different expressed genes named Data set Ⅱ were identified through lab data analysis platform named GCBI,Data set Ⅰ and Ⅱ were intersected,then pathways and Internal relations between these genes were explored through three methods include gene oncology(GO),KEGG pathway and protein-protein interaction analysis(PPI).RESULTS 455 different expressed genes were got from 3 gene expression data sets met the inclusion criteria,Herbal ingredients from HeiGuTeng ZhuiFeng HuoLuo capsule and 432 targeting genes were got from BATMAN-TCM Database,then got intersected again,27 potential target genes for HeiGuTeng ZhuiFeng HuoLuo capsule acting on Rheumatoid Arthritis came out.Next,bioinformatics analysis and references showed 5 genes(STAT1、MAPK1、RAC2、CDK1、PRKCB) get a close relationship and these genes were enriched in signaling pathway relative to STAT1-ERKs pathway.These suggested that HeiGuTeng ZhuiFeng HuoLuo capsule may be effect on RA via regulatting STAT1-ERKs in synoviocytes.CONCLUSION Bioinformatics analysis may be helpful to research RA gene microarray data and predict treating target genes and signaling pathways of Miao medicine HeiGuTeng ZhuiFeng HuoLuo capsule acting on Rheumatoid Arthritis.
