摘 要:目的建立利用UPLC-MS/MS法测定人血浆罗哌卡因的分析方法。方法血浆样品经甲醇直接沉淀,固定相为Acquity UPLC BEH C18(2.1mm×50mm,1.7μm),流动相为0.02%甲酸水(B) 60%~甲醇(C) 40%,流速0.2mL·min^(-1),柱温:30℃,质谱条件为电喷雾离子源、正离子方式、选择反应监测(SRM),罗哌卡因离子反应分别为m/z:275.4→126。结果罗哌卡因血浆浓度在50~1000ng·mL-1(r2=0.998,n=6)内线性关系良好,萃取回收率在77%~84%之间,回收率在93%~100%之间(RSD<1%,n=5),日间、日内RSD均<1%(n=5),最低检测限为10ng·mL-1(S/N≥10)。结论建立的检测方法选择性强、灵敏度高、重现性好,可用于测定人体血浆中罗派卡因的血药浓度。OBJECTIVE To establish a method for the determination of ropivacaine in human plasma through UPLC-MS/MS.METHODS The samples of plasma were precipitated directly by methanol.The stationary phase was Acquity UPLC BEH C18(2.1 mm×50 mm,1.7μm).The flow phase was 0.02% formate water(B) 60%-methanol(C) 40%,flow rate was 0.2 mL·min-1,column temperature was 30℃,mass spectrometry conditions were electrospray ion source,positive ion mode and selective reaction monitoring(SRM).The ionic response of ropivacaine was m/z:275.4 to 126,respectively.RESULTS The linear relationship between the plasma concentration of ropivacaine in 50~1000 ng·mL-1(r2= 0.998,n = 6) was good,the recovery rate was from 77% to 84%,the recovery rate was from 93% to 100%(RSD<1%,n = 5).with the intra-day and inter-day RSDs were both less than<1%(n = 5),and the minimum detection limit was 10 ng·mL-1(S/N > 10).RESULTS This method is highly selective,sensitive and reproducible,and could be used to determine the plasma concentration of ropivacaine in human plasma.
