摘 要:目的:探讨心康冲剂对慢性心衰大鼠PTEN/AkT/MMP-2蛋白的干预作用。方法:SD大鼠以阿霉素按1.5mg/kg体质量腹腔注射复制慢性心衰模型,模型成功后各组给予相应药物处理。心脏切片行Masson染色并计算胶原容积分数(CVF),采用Real-time PCR法检测PTEN mRNA表达水平,Western blot法检测心肌组织PTEN、AkT、p-AkT、MMP-2蛋白质表达水平。结果:与正常组比较,模型组大鼠心肌纤维被银蓝色胶原纤维组织取代,CVF比值明显增大,心肌PTENmRNA表达下降,PTEN蛋白表达下降,AkT、p-AkT、MMP-2蛋白表达升高。与模型组比较,心康低、中、高剂组纤维化相对减轻,CVF显著下降,PTENmRNA表达升高,PTEN蛋白表达升高,AkT、p-AkT、MMP-2表达均下降;与对照组比较,心康高剂组PTEN表达减少而MMP-2升高,余心康冲剂低、中、高剂量组CVF、AkT、MMP-2、PTENmRNA比较差异无统计学意义。结论:心康冲剂抗心肌纤维化可能与增加PTEN表达、下调AkT、MMP-2表达有关,与芪苈强心胶囊疗效相当。Objective:To investigate the intervention effect of Xin Kang Granule(XKG) on PTEN/AkT/MMP-2 pathway in rats with heart failure.Method: SD rats were injected intraperitoneally with adriamycin at 1.5mg/kg weight. After the model was successfully treated with corresponding drugs in each group,heart slices were stained by Masson and calculated the collagen volume fraction(CVF),mRNA expression level of PTEN was detected by Real-time PCR,Western blot was used to detect protein expression level of PTEN,AkT,p-AkT and MMP-2 in myocardial tissue of rats.Results: Compared with the normal group,rats in the model group showed the muscle fibers being replaced by blue collagen fibers,CVF increased significantly,the mRNA and protein expression level of PTEN down-regulated,protein expression level of p-AkT,,AKT and MMP-2 up-regulated.but PTEN less.Compared with model group,rats in low,middle and high dose XKG groups showed blue collagen fibrous tissue decreased,CVF decreased significantly,the mRNA and protein expression level of PTEN up-regulated,protein expression level of p-AkT,AKT and MMP-2 down-regulated.,but PTEN more.Compared with the control group,rats in high dose group showed the expression of PTEN reduced but MMP-2 increased,but rats in low,middle dose XKG groups showed no significant difference in CVF,p-AkT,AkT,MMP-2,PTENmRNA.Conclusion:XKG has effect of anti myocardial fibrosis may be related to the increase of PTEN expression and down regulation of AkT and MMP-2 expression.
